HPV整合靶基因INHBA与宫颈癌患者预后的综合性分析

摘要 目的利用生物信息学和细胞学实验探究INHBA基因在宫颈癌中的临床意义和相关机制。方法利用TCGA、GEO等数据库检测INHBA在宫颈癌样本中的表达变化;利用GEPI2A数据库分析INHBA表达与宫颈癌患者预后的相关性;利用LinkedOmics数据库挖掘宫颈癌中与INHBA共表达的基因并进行GO和KEGG信号通路分析;利用ENCORI数据库探究INHBA的上游调控miRNA,并在CancerMIRNome数据库中验证相关miRNA在宫颈癌中的表达。在宫颈癌细胞HeLa和SiHa中敲低INHBA表达,进行CCK-8、细胞划痕和Transwell小室实验,探究INHBA对细胞增殖和迁移的影响;结果与正常样本相比,INHBA基因在宫颈癌样本中高表达,并且与患者的不良预后相关。miR-532-5p可能是INHBA的上游调控分子。INHBA能够通过调控细胞增殖和迁移影响癌症进展。结论INHBA表达升高是宫颈癌患者不良预后的潜在指标。 Objective To explore the clinical significance and related mechanism of INHBA gene in cervical cancer by bioinformatics method.Methods The expression of INHBA in cervical cancer samples was detected by TCGA,GEPIA and GEO databases;The correlation between INHBA expression and prognosis of cervical cancer patients was analyzed by GEPIA2 database;LinkedOmics databases was used to obtain the list of genes co-expressed with INHBA in cervical cancer.GO and KEGG signal pathways were also analyzed;ENCORI database was used to explore the upstream regulatory miRNA of INHBA,and to verify the prognostic diagnostic value of relevant miRNA in CancerMIRNome database;We knocked down the expression of INHBA in cervical cancer cells HeLa and SiHa,and conducted CCK-8,cell scratch,and transwell cell experiments to investigate the effect of INHBA on cell proliferation and migration.Results Compared with normal samples,INHBA gene was highly expressed in cervical cancer samples and was associated with poor prognosis.miR-532-5p could be the key upstream regulatory molecules of INHBA.INHBA could regulate cancer progression by affecting cell proliferation and migration.Conclusion NHBA is a potential indicator of poor prognosis in patients with cervical cancer.
