细胞分裂周期蛋白42、性别决定区Y框蛋白2在骨肉瘤组织和骨软骨瘤组织的表达及其临床意义

摘要 目的检测细胞分裂周期蛋白42(Cdc42)和性别决定区Y框蛋白2(SOX2)在骨软骨瘤组织、骨肉瘤组织中的表达, 探讨Cdc42和SOX2与骨肉瘤临床病理因素及预后之间的关系。方法采用免疫组织化学法检测2018年3月至2022年11月青岛市中医医院病理确诊的96例骨肉瘤组织和35例骨软骨瘤组织中Cdc42和SOX2的表达, 采用χ^(2)检验进行统计学分析。结果 Cdc42在骨肉瘤组织中阳性表达率为62.50%(60/96), 明显高于骨软骨瘤组织中阳性表达率[11.43%(4/35)], 差异有统计学意义(χ^(2)=26.774, P<0.01)。Cdc42表达水平与骨肉瘤患者恩内金分期(Eneeking分期)、肺转移明显相关(χ^(2)=5.039、5.248, P<0.05), Cdc42表达水平与骨肉瘤患者病变部位、性别、组织类型、年龄、软组织浸润、ALP水平无相关(χ^(2)=0.012、0.183、0.027、0.028、0.200、0.072, P>0.05)。SOX2在骨肉瘤组织中阳性表达率为42.71%(41/96), 明显高于骨软骨瘤组织中阳性表达率[8.57%(3/35)], 差异有统计学意义(χ^(2)=13.399, P<0.01)。SOX2表达水平与骨肉瘤患者Eneeking分期、软组织浸润、肺转移明显相关(χ^(2)=4.690、5.243、6.152, P<0.05), SOX2表达水平与骨肉瘤患者病变部位、性别、组织类型、年龄、ALP水平无相关(χ^(2)=0.051、0.001、0.083、0.002、0.319, P>0.05)。结论 Cdc42和SOX2在骨肉瘤组织中表达上调, 联合检测Cdc42和SOX2有助于判断骨肉瘤生物学行为和预后。 Objective The expression of cell division cycle 42(Cdc42)and sex determining re-gion Y-box 2(SOX2)in osteochondromas and osteosarcoma tissues wase detected.To investigate the rela-tionship between Cdc42,SOX2 and the clinicopathological factors and prognosis of osteosarcoma.Methods Immunohistochemistry was used to detect the expression of Cdc42 and SOx2 in 96 cases of osteosarcoma tissues and 35 cases of osteochondroma tissues confirmed by pathology in Qingdao Hospital of Traditional Chinese Medicine from March 2018 to November 2022.Chi-square test was used for statistical analysis.Results The positive expression rate of Cdc42 in osteosarcoma tissues was 62.50%(60/96),which was significantly higher than that in osteochondroma tissues[11.43%(4/35),χ^(2)=26.774,P<0.01].The expression level of Cdc42 was significantly correlated with Eneeking stage and lung metastasis in osteosarco-ma patients(Respectively,χ^(2)=5.039,5.248,P<0.05),but not significantly correlated with lesion location,gender,tissue type,age,soft tissue infiltration and ALP level in osteosarcoma patients(respectively,χ^(2)=0.012,0.183,0.027,0.028,0.200,0.072,P>0.05).The positive expression rate of SOX2 in osteosarcoma tissues was 42.71%(41/96),which was significantly higher than that in osteochondroma tissues[8.57%(3/35),χ^(2)=13.399,P<0.01].The expression level of SOX2 was significantly correlated with Eneeking stage,soft tissue invasion and lung metastasis of osteosarcoma patients(respectively.χ^(2)=4.690,5.243,6.152,P<0.05),but not with lesion loeation,gender,histological type,age and ALP level in patients with osteosarcoma(respectively,χ^(2)=0.051,0.001,0.083,0.002,0.319,P>0.05).Conclusion Cdc42 and SOX2 are up-regulated in osteosarcoma tissues.Combined detection of Cdc42 and SOX2 may help to determine the biological behavior and prognosis of osteosarcoma.
