杂交兰‘红美人’×寒兰‘杨贵妃’杂交后代增殖与分化研究 被引量:1

Study on the Proliferation and Differentiation of Rhizomes in the Hybrid Progeny of Hybrid Orchid‘Hongmeiren’X Cymbidium kanran‘Yangguifei'

摘要 以杂交兰‘红美人’×寒兰‘杨贵妃’杂交后代的根状茎为试材,采用L9(34)正交实验方法,研究了6-BA、NAA、TDZ、不同光周期及不同基本培养基对根状茎增殖、叶芽分化以及花芽分化的影响,以期探究杂交兰根状茎增殖与分化适合的条件。结果表明:在1.0 g·L^(-1)花宝1号+1.0 g·L^(-1)花宝2号+1.0 mg·L^(-1)TDZ+1.0 mg·L^(-1)NAA+0.1 g·L^(-1)活性炭+40.0 g·L^(-1)蔗糖+7.0 g·L^(-1)琼脂+6.0 g·L^(-1)蛋白胨+40.0 g·L^(-1)香蕉+40.0 g·L^(-1)马铃薯处理中,根状茎的增殖效果最好;光周期12 h/12 h(光照/黑暗)的增殖系数最高,10 h/14 h(光照/黑暗)的生长速度最快;在2.0 g·L^(-1)花宝1号+1.0 g·L^(-1)花宝2号+5.0 mg·L^(-1)6-BA+1.0 mg·L^(-1)NAA+0.1 g·L^(-1)活性炭+40.0 g·L^(-1)蔗糖+7.0 g·L^(-1)琼脂+6.0 g·L^(-1)蛋白胨+50.0 mg·L^(-1)椰汁的处理中,叶芽分化率最优;在基本培养基MS+5.0 mg·L^(-1)6-BA+1.0 mg·L^(-1)NAA+0.1 g·L^(-1)活性炭+40.0 g·L^(-1)蔗糖+7.0 g·L^(-1)琼脂+6.0 g·L^(-1)蛋白胨+40.0 g·L^(-1)香蕉+40.0 g·L^(-1)马铃薯的处理中,根状茎产生了花芽分化。 Taking hybrid orchid‘Hongmeiren'X Cymbidium kanran‘Yangguifei'of rhizomes as experimental material,using Lg(3*)orthogonal experiment method,the effects of 6-BA,NAA,TDZ on different photoperiod and basic culture medium of rhizomes proliferation,leaf bud differentiation and flower bud differentiation were studied,in order to explore the hybrid laingen stems proliferation and differentiation of suitable conditions.The results showed that the best formula of rhizome proliferation,2.0 g·L^(-1)Huabao 1+1.0 g·L^(-1)Huabao 2+1.0 mg·L^(-1)TDZ+1.0 mg·L^(-1)NAA+0.1 g·L^(-1)activated carbon+40.0 g·L^(-1)sucrose+7.0 g·L^(-1)agar+6.0 g·L^(-1)proteinpeptone+40.0 g·L^(-1)banana+40.0 g·L-'potato;the proliferation coefficient was highest at 12 hours/12 hours(light/dark)and the fastest growth rate at 10 hours/14 hours(light/dark).The best formula of leaf bud differentiation,2.0 g·L^(-1)Huabao 1+1.0 g·L^(-1)Huabao 2+5.0 mg·L^(-1)6-BA+1.0 mg·L^(-1)NAA+0.1 g·L^(-1)activated carbon+40.0 g·L^(-1)sucrose+7.0 g·L^(-1)agar+6.0 g·L-'peptone+50.0 mg·L^(-1)coconut milk.In MS+5.0 mg·L^(-1)6-BA+1.0 mg·L^(-1)NAA+0.1 g·L^(-1)activated carbon+40.0 g·L^(-1)sucrose+7.0 g·L^(-1)agar+6.0 g·L-I proteinpeptone+40.0 g·L^(-1)banana+40.0 g·L^(-1)potato,the rhizome produced flower buds.
