黄连素通过调控丝裂原活化蛋白激酶信号通路治疗各科疾病的药理机制研究进展

Research progress on the pharmacological mechanism of berberine in the treatment of various diseases based on MAPK signaling pathway

黄连素是广泛存在于黄连、黄柏、小檗等中草药的一种异喹啉生物碱,具有多种药理作用。黄连素发挥多种药理机制与丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)信号通路有关。本文发现黄连素通过调控MAPK信号通路发挥以下作用:抑制癌细胞转移、增殖,增加耐药癌细胞敏感性发挥抗癌作用;抑制病毒复制,抑制细菌诱导的炎症,抑制真菌增殖发挥抗病原体作用;促进组织葡萄糖摄取降血糖,改善胰岛素抵抗,保护胰岛β细胞发挥抗糖尿病作用;抑制血管平滑肌细胞增殖,抑制血管内皮细胞凋亡,降血脂发挥抗动脉粥样硬化作用;抑制心肌缺血,抑制心肌重塑发挥心脏保护作用;抑制β淀粉样蛋白表达,抑制神经炎症,抑制神经细胞死亡,改善糖尿病性神经病变发挥神经系统保护作用;抗胃肠炎,抗胰腺炎,肝保护作用发挥消化系统保护作用;抗糖尿病肾病,抗药物性肾损伤,抗肾缺血发挥肾脏保护作用;抗过敏反应,抗类风湿关节炎,抗骨关节炎,抗牙周炎、鼻息肉等发挥抗炎作用。

Berberine is an isoquinoline alkaloid widely existed in Chinese herbal medicine,such as Coptis chinensis,Cortex Phellodendri and Berberis vulgaris.Berberine has a variety of pharmacological effects.The various pharmacological mechanisms of berberine are related to the mitogen-activated protein kinase(MAPK)signaling pathway.Berberine plays the following roles by regulating MAPK signaling pathway.Berberine plays an anticancer role by inhibiting the metastasis and proliferation of cancer cells and increasing the sensitivity of drug-resistant cancer cells.Berberine plays an anti-pathogen role by inhibiting viral replication,inhibiting bacteria-induced inflammation,and inhibiting fungal proliferation.Berberine plays an anti-diabetic role by promoting tissue glucose uptake,lowering blood glucose,improving insulin resistance,and protecting isletβ-cells.Berberine plays an anti-atherosclerosis role by inhibiting vascular smooth muscle cell proliferation,inhibiting vascular endothelial cell apoptosis and lowering blood lipids.Berberine plays a cardiac protective role by inhibiting myocardial ischemia and myocardial remodeling.Berberine plays a protective role in the nervous system by inhibiting the expression of amyloid beta protein,inhibiting neuroinflammation,nerve cell death,and improving diabetic neuropathy.Berberine plays a protective role in digestive system through anti-gastroenteritis,anti-pancreatitis and liver protection.Berberine plays a renal protective role by anti-diabetic nephropathy,anti-drug induced renal injury and anti-renal ischemia.Berberine has anti-inflammatory effects through anti-allergic reactions,anti-rheumatoid arthritis,anti-osteoarthritis,anti-periodontitis,nasal polyps and so on.
