益艾康对HIV-1感染者CD_(4)^(+)T淋巴细胞ICAM-1、LFA-1表达水平的影响 被引量:1

Effects of Yiai Kang on ICAM-1 and LFA-1 expression levels of CD_(4)^(+)T lymphocytes in HIV-1 infected patients

摘要 目的:观察益艾康对人类免疫缺陷病毒(HIV)-1感染者CD_(4)^(+)T淋巴细胞ICAM-1、LFA-1表达水平的影响。方法:收集20例HIV-1感染者(HARRT治疗组和益艾康+HARRT治疗组各10例)和6名健康组EDTA抗凝全血,流式细胞技术检测各组CD_(4)^(+)T淋巴细胞数量,ELISA检测血清中ICAM-1、LFA-1α水平,荧光定量PCR方法观察各组ICAM-1、LFA-1mRNA表达水平。结果:与健康组比较,两治疗组CD_(4)^(+)T淋巴细胞绝对计数显著降低(P<0.05),HARRT治疗组ICAM-1、LFA-1 mRNA表达显著升高(P<0.05);与HARRT治疗组比较,艾益康+HAART治疗组CD_(4)^(+)T淋巴细胞绝对计数显著升高(P<0.05),ICAM-1水平及LFA-1mRNA表达显著降低(P<0.05)。结论:益艾康可降低HIV-1感染者CD_(4)^(+)T淋巴细胞ICAM-1、LFA-1的表达,益艾康可能通过稳定或提高CD_(4)^(+)T淋巴细胞数量来调节HIV-1患者的免疫功能。 Objective:To observe the effects of Yiai Kang on the expression levels of intercellular cell adhesion molecule-1(ICAM-1)and lymphocyte function-associated antigen(LFA)-1 in CD_(4)^(+)T lymphocytes infected with human immunodeficiency virus(HIV)-1.Methods:EDTA anticoagulated whole blood was collected from 20 HIV-1 infected patients(10 cases in HARRT treatment group and 10 cases in Yiai Kang+HARRT treatment group)and 6 healthy people.The number of CD_(4)^(+)T lymphocytes in each group was detected by flow cytometry,the levels of ICAM-1 and LFA-1αin serum were detected by ELISA,and the expression levels of ICAM-1 and LFA-1 mRNA in each group were observed by fluorescence quantitative PCR.Results:Compared with the healthy group,the absolute count of CD_(4)^(+)T lymphocytes in the two treatment groups decreased significantly(P<0.05),and the mRNA expressions of ICAM-1 and LFA-1 in the HARRT treatment group increased significantly(P<0.05).Compared with the HARRT treatment group,the absolute CD_(4)^(+)T lymphocyte count in the Yiai Kang+HAART treatment group increased significantly(P<0.05),while the ICAM-1 level and LFA-1 mRNA expression decreased significantly(P<0.05).Conclusion:Yiai Kang can reduce the expression of ICAM-1 and LFA-1 in CD_(4)^(+)T lymphocytes of HIV-1 infected patients,Yiai Kang may regulate the immune function of HIV-1 patients by stabilizing or increasing the number of CD_(4)^(+)T lymphocytes.
