hsa-miR-1-3p、FN1、DKK1在子痫前期中的表达及相关性研究

Expression and correlation of hsa-miR-1-3p,FN1 and DKK1 in pre-eclampsia

摘要 目的 探讨hsa-miR-1-3p、FN1、DKK1在子痫前期中的表达及意义,分析其潜在相关性和临床应用价值。方法 选取子痫前期表达谱芯片GSE30186和GSE44711数据集,生物信息学分析hsa-miR-1-3p、FN1、DKK1的表达情况和相关性,并经realtime PCR和免疫组化初步检验其相关性。结果 FN1和DKK1 mRNA在表达谱芯片和收集的子痫前期组织中相对于正常组织均高表达(P<0.05),FN1和DKK1蛋白表达在子痫组织中相较于正常组织具有更高水平的表达。hsa-miR-1-3p在子痫前期组织中表达水平低于正常对照组织(P<0.05)。生物信息学分析预测hsa-miR-1-3p居于连接FN1和DKK1的关键位置。结论 FN1和DKK1在子痫前期中高表达,hsa-miR-1-3p可能是潜在调控FN1和DKK1表达水平的关键因素,hsa-miR-1-3p、FN1、DKK1是子痫前期患者中的有效生物标志物。 Objective To investigate the expression and significance of hsa-miR-1-3p,FN1 and DKK1 in pre-eclampsia and to analyse their potential relevance and clinical application.Methods The GSE30186 and GSE44711 datasets of preeclampsia expression profiling microarrays were selected,and the expression and correlation of hsa-miR-1-3p,FN1,DKK1 were bioinformatically analyzed and verified by realtime PCR and immunochemistry.Results FN1 and DKK1 mRNAs were highly expressed in both expression profiling microarrays and collected pre-eclamptic tissues relative to normal tissues(P<0.05),and FN1 and DKK1 protein expression was higher in pre-eclamptic tissues compared to normal tissues.hsa-miR-1-3p was expressed at lower levels in pre-eclamptic tissues than in normal control tissues(P<0.05).Bioinformatics analysis predicts that hsa-miR-1-3p resides in a key position linking FN1 and DKK1.Conclusion FN1 and DKK1 are highly expressed in pre-eclampsia.hsa-miR-1-3p may be a key factor regulating the expression levels of FN1 and DKK1.hsa-miR-1-3p,FN1,and DKK1 are potent biomarkers in pre-eclampsia.
