甘西鼠尾草总酚酸提取物对电离辐射大鼠急性放射性肠炎的保护作用

Protective effect of Salvia prezewalskii extract of total phenolic acids on acute radiation induced bowel injury after ionizing radiation in rats

摘要 目的:研究甘西鼠尾草总酚酸提取物(Salvia prezewalskii extract of total phenolic acids,SPE)对电离辐射大鼠急性放射性肠炎损伤的保护作用。方法:60只SD雄性大鼠随机分为6组,每组10只。除正常对照组不予辐射外,其余各组大鼠均经麻醉后接受10 Gy高能X射线单次腹部照射1次,建立大鼠急性放射性肠炎模型。辐射后,SPE各组大鼠以20,40,80 mg·kg^(-1)剂量分别灌胃给予SPE混悬溶液,1次/天,连续给药7 d。于辐射前及辐射后第1~7天观察各组大鼠体征并称定体质量。第7天处死大鼠后,光学显微镜下观察结肠组织病理,采用ELISA试剂盒检测外周血清炎性因子白细胞介素-1β(interleukin-1β,IL-1β)、IL-6、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)含量及结肠组织髓过氧化物酶(myeloperoxidase,MPO)含量,采用蛋白印迹法检测结肠组织炎症小体NLRP3及凋亡相关蛋白Bcl-2、Bax、caspase-3、Cyto-C表达。结果:治疗后,SPE可显著增加辐射损伤大鼠体质量,改善结肠组织病理状态,降低血清IL-1β、IL-6、TNF-α含量及结肠组织MPO含量,同时抑制结肠组织NLRP3及Bax、caspase-3、Cyto-C蛋白表达。结论:SPE对电离辐射大鼠急性放射性肠炎具有保护作用。 OBJECTIVE To investigate protective effect of Salvia prezewalskii extract of total phenolic acids(SPE)on acute radiation induced bowel injury after ionizing radiation in rats.METHODS Sixty SD male rats were randomly divided into 6 groups,with 10 rats in each group.Except normal control group,rats in other groups were received 10 Gy X-ray single abdominal radiation once after anesthesia to establish acute radiation induced bowel injury model of rats.After radiation,SPE suspension solution was administered intragastrically to each SPE group rats at doses of 20,40 and 80 mg·kg^(-1) for 7 consecutive days respectively at 24 h intervals.The physical signs in each group rats were observed and the body mass were weighed on one day before,day 1 and day 7 after radiation,respectively.After the rats were harvested on day 7,the colon histopathology was observed under light microscope.The content of inflammatory factor interleukin-1β(IL-1β),IL-6 and tumor necrosis factor-α(TNF-α)in peripheral serum and the content of myeloperoxidase(MPO)in colon tissue were detected with ELISA.The expression of inflammasome NLRP3,and apoptosis related protein Bcl-2,Bax,caspase-3 and Cyto-C in colon tissue were detected by Western blot.RESULTS After treatment,SPE could significantly increase the body mass of radiation injured rats,improve the pathological state of colon tissue,reduce IL-1β,IL-6 and TNF-αcontents in serum and MPO content in colon tissue,and inhibit NLRP3,Bax,caspase-3 and Cyto-C expression in colon tissue.CONCLUSION SPE has protective effect on acute radiation induced bowel injury after ionizing radiation in rats.
