摘要
【目的】为了减少猕猴桃溃疡病植物源抗菌药剂的原材料的生产成本,使中药材的利用率最大化,并开发适合针对猕猴桃溃疡病菌的植物源复配抑菌剂。【方法】通过测量不同提取条件下的石菖蒲提取物对猕猴桃溃疡病菌的抑菌圈直径,得到石菖蒲的较佳提取工艺,并用响应面分析法筛选出石菖蒲、黄芩、丁香、广藿香、大蒜和牡丹6种中药材组成的最佳复配物。【结果】石菖蒲提取物的较佳工艺条件为:乙醇浓度81.171%、提取温度54.730℃、料液比1∶25、提取温度50℃、超声功率700W,此条件下的抑菌圈直径可达27.81mm,复配抑菌剂的最佳配方工艺为:黄芩∶石菖蒲∶大蒜∶广藿香∶牡丹∶丁香=182.310∶306.104∶521.432∶453.106∶153.199∶362.565,在此条件下,1.0g/mL的复配物实际试验测得的抑制圈直径可达33.860mm。【结论】确定了石菖蒲提取物对猕猴桃溃疡病菌产生抑菌作用的较佳提取工艺,且同条件下,复配物有更强的抑菌作用。
【Objective】In order to reduce the production cost of plant-derived antibacterial agents for kiwifruit ulcer disease,maximize the utilization rate of Chinese medicinal materials,and develop plantderived compound antibacterial agents suitable for kiwifruit ulcer disease.【Method】By measuring the diameters of the inhibition zone of Acorus tatarinowii extract under different extraction conditions against Pseudomonas syringae,the better extraction technology of Acorus tatarinowii was obtained.The optimal compound of Acorus tatarinowii,Scutellaria baicalensis,Syzygium aromaticum,Pogostemon cablin,Al⁃lium sativum and paeonia suffruticosa was screened by response surface analysis(RSM).【Result】The better technological conditions of Acorus tatarinowii extract were as follows:ethanol concentration of 81.171%,extraction temperature of 54.730℃,solid-liquid ratio of 1∶25,extraction temperature of 50℃,ultrasonic power of 700 W.Under these conditions,the diameter of the inhibition zone could reach 27.81 mm.The optimum formula of compound antibacterial agent was:Scutellaria baicalensis∶Scutellaria ba⁃icalensis∶Allium sativum∶Pogostemon cablin∶Paeonia suffruticosa∶Syzygium aromaticum=182.310∶306.104∶521.432∶453.106∶153.199∶362.565.Under these conditions,the diameter of inhibition ring measured by the 1.0 g/mL compound could reach 33.860 mm in the actual test.【Conclusion】The optimal extraction process of Acorus tatarinowii extract against kiwifruit ulcer disease was determined,and the compound had stronger antibacterial effect under the same conditions.
