摘要
目的探究人乳头状甲状腺癌(papillary thyroid cancer,PTC)细胞TPC-1中长链非编码RNA(long non-coding RNA,lncRNA)肺腺癌转移相关转录本1(metastasis associated in lung adenocarcinoma transcript 1,MALAT1)的表达,及沉默MALAT1对TPC-1细胞增殖、侵袭及迁移的影响。方法体外培养PTC细胞系TPC-1和人正常甲状腺滤泡上皮细胞系Nthy-ori3-1,采用定量反转录PCR(quantitative reverse transcriptase-mediated PCR,qRT-PCR)检测MALAT1在TPC-1和Nthy-ori3-1细胞系中的表达水平;构建干扰小RNA(small interfering RNA,siRNA),取对数生长期的TPC-1细胞,经脂质体瞬时转染技术干扰TPC-1细胞中MALAT1的表达,设置对照组(仅加入培养基)、MALAT1沉默对照组(si-NC组,加入空质粒载体)、MALAT1沉默组(si-MALAT1组,加入MALAT1-siRNA质粒),并采用qRT-PCR检测MALAT1的mRNA表达情况;CCK-8检测TPC-1细胞的增殖情况;Transwell小室实验检测TPC-1细胞侵袭能力;划痕试验检测TPC-1细胞迁移能力。结果与Nthy-ori3-1细胞相比,TPC-1中MALAT1的表达上调(P<0.01)。与对照组、si-NC组比较,si-MALAT1组MALAT1表达水平明显降低(均P<0.01),TPC-1细胞增殖、侵袭及迁移能力均显著减弱(P<0.001,P<0.01,P<0.05)。结论沉默MALAT1可抑制人PTC细胞TPC-1的增殖、侵袭及迁移能力。
Objective To investigate the expression of long non-coding RNA(lncRNA)metastasis associated in lung adenocarcinoma transcript 1(MALAT1)in human papillary thyroid cancer(PTC)cells TPC-1,and the effect of silencing MALAT1 on TPC-1 cell proliferation,invasion,and migration.Methods In vitro cultivation of PTC cell line TPC-1 and human normal thyroid follicular epithelial cell line Nthy-ori3-1,quantitative reverse transcriptase-mediated PCR(qRT-PCR)was used to detect the expression level of MALAT1 in TPC-1 and Nthy-ori3-1 cell lines;Construct small interfering RNA(siRNA),take logarithmic growth stage TPC-1 cells,and use liposome transient transfection technology to interfere with the expression of MALAT1 in TPC-1 cells.Set up a control group(only adding culture medium),a MALAT1 silencing control group(si-NC group,adding empty plasmid carrier),and a MALAT1 silencing group(si-MALAT1 group,adding MALAT1-siRNA plasmid),and use qRT-PCR to detect the mRNA expression of MALAT1;CCK-8 was used to detect the proliferation of TPC-1 cells;Transwell cell assay was used to detect the invasiveness of TPC-1 cells;The ability of TPC-1 Cell migration was tested by scratch test.Results Compared with Nthy-ori3-1 cell,the MALAT1 expression in TPC-1 cell was increased(P<0.01).Compared with control group and si-NC group,respectively,the expression of MALAT1 was reduced(both P<0.01),the proliferation ability,invasion and migration ability of TPC-1 cell in the si-MALAT1 group were reduced(P<0.001,P<0.01,P<0.05).Conclusion Silencing MALAT1 can inhibit the proliferation,invasion and migration of TPC-1 cell.
