摘要
以苯乙烯为单体、甲基丙烯酸缩水甘油酯和1-乙烯基-3-丁基-咪唑溴盐为共聚物,通过分散聚合法合成了固定化酶载体。通过静电相互作用和共价结合两种作用力共同对脂肪酶进行固定化,得到脂肪酶-聚离子液体-甲基丙烯酸缩水甘油酯-苯乙烯微球(lipase@PIGSs)。利用XRD、SEM和FT-IR等技术对lipase@PIGSs进行表征,并对其稳定性、重复利用性等酶学性质与游离脂肪酶进行了比较。结果表明,固定化后的脂肪酶稳定性和重复利用性均有显著提高,载酶量达到38.0mg/g,酶活力达到9.90U/mg,高于游离酶酶活力(9.39U/mg)。酶促反应动力学结果表明,lipase@PIGSs具有良好的催化活性。催化应用实验利用lipase@PIGSs成功催化了共轭亚油酸乙酯的合成。
Using styrene as monomers,glycidyl methacrylate and 1-vinyl-3-butyl imidazolium bromide([VBIm]Br)as copolymers,the immobilized enzyme carriers were synthesized by dispersion polymerization.Lipase-poly(ionic liquid)-glycidyl methacrylate-styrene microspheres(lipase@PIGSs)were obtained by immobilization of lipase through electrostatic interaction and covalent binding.The lipase@PIGSs were characterized by XRD,SEM and FT-IR,and their enzymatic properties such as stability and reusability were compared with those of the free lipase.The results showed that the stability and reusability of the immobilized lipase were significantly improved,with the enzyme loading of 38.0 mg/g and enzyme activity of 9.90 U/mg,which was higher than that of the free lipase(9.39 U/mg).The kinetic parameters of the enzymatic reaction indicated that lipase@PIGSs had good catalytic activity.
