肝细胞癌关键基因分析及相关免疫机制分析

摘要 目的筛选肝细胞癌关键致病基因,分析免疫相关致病机制。方法GEO数据库下载芯片数据,筛选差异基因,并进行注释。构建差异基因的蛋白质相互作用(protain-protain interaction,PPI),计算核心基因。分析hub基因对预后的影响,并探索免疫学机制。结果通过GEO2R的分析筛选并以韦恩图取交集得到差异基因798个。利用Cytoscape软件计算得到核心关键基因共6个,分别为CDK1、CCNA2、CCNB1、CDC20、TOP2A、KIF11,上述基因的高表达会降低患者生存率,可能是相应蛋白会抑制免疫细胞并辅助肿瘤细胞进行免疫逃逸。结论本研究找到影响肝细胞癌发生发展的关键蛋白,分析其促进肿瘤发展的相关免疫机制,为后续临床试验及药物研发提供理论基础。 Objective To screen the key pathogenic genes of hepatocellular carcinoma,and analyze the immunerelated pathogenic mechanisms.Methods The microarray data was downloaded from the GEO database,and the differ⁃ential genes were screened and annotated.Construct the protain-protain interaction(PPI)of differential genes and cal⁃culate the core genes.Analyze the effect of hub gene on prognosis and explore the immunological mechanism.Results Through GEO2R analysis and screening,798 differential genes were obtained through the intersection of Venn dia⁃grams.6 core key genes were calculated by using Cytoscape software,namely CDK1,CCNA2,CCNB1,CDC20,TOP2A,KIF11.The high expression of the above genes will reduce the survival rate of patients,possibly because the corresponding proteins can inhibit immune cells and assist tumor cells in immune escape.Conclusion This study finds the key proteins that affect the occurrence and development of hepatocellular carcinoma,and analyzes the re⁃lated immune mechanisms that promote tumor development to provide a theoretical basis for subsequent clinical trials and drug development.
