摘要
目的:建立电感耦合等离子体质谱法(ICP-MS)测定不同产地黄精中砷、铅、铬、镉、汞含量的方法,进行含量分析,为黄精的医用和食疗安全提供理论基础。方法:采用ICP-MS法对52份野生黄精样品中5种重金属进行检测,内标法定量,并进行数据分析。结果:野生黄精中5种重金属的方法检出限为0.002~0.020 mg/kg,回收率为95.5%~104.4%,精密度为2.8%~4.6%;野生黄精根茎中含量高低依次为铬、铅、镉、砷和汞,各元素间呈正相关;巫山的铬和砷显著高于其他地区,奉节的铅最高,石柱的镉显著高于其他地区;黄精不同部位的重金属含量高低:根须>根茎>叶子>种子,其中根须的铬含量高达4.406±1.021 mg/kg。结论:野生黄精的重金属含量存在一定的地域性差异;不同部位的含量也有差异:地下部位(根须、根茎)含量大于地上部位(叶子、种子)。本研究可为黄精的外源性污染提供数据支撑。
Objective:To establish a method for the determination of arsenic,lead,chromium,cadmium,and mercury in Polygonatum sibiricum Red.from different producing areas by Inductively Coupled Plasma Mass Spectrometry(ICP-MS),and to analyze the content of samples,provided a theoretical basis for the safety of medicine and food therapy of P.sibiricum.Methods:Five heavy metals in 52 samples were detected by ICP-MS method and quantified by internal standard method,and data were analyzed.Results:The detection limit of heavy metals in wild P.sibiricum were 0.002-0.20 mg/kg,the recovery rate of standard addition were 95.5%-104.4%,and the precision were 2.8%-4.6%.The contents of 5 heavy metals in rhizome of P.sibiricum were:chromium,lead,cadmium,arsenic and mercury in turn,and there was a positive correlation between the elements.Chromium and arsenic in Wushan were significantly higher than those in other areas,lead in Fengjie was the highest,and cadmium in Shizhu was significantly higher than others.The contents of heavy metals in different parts of P.sibiricum were:root,rhizome,leaf,seed in turn,and the chromium content in root was 4.406±1.021 mg/kg.Conclusion:there were some regional differences in the content of heavy metals in wild P.sibiricum;there were also differences in the content of different parts.The content of heavy metals in underground parts(roots,rhizomes)was greater than that in aboveground parts(leaves,seeds).This study could provide data support for the exogenous pollution of P.sibiricum.
