摘要
研究旨在了解KISS1基因对猪繁殖力的影响,以78头初产柯乐猪为材料,采用聚合酶链式反应(PCR)产物直接测序与人工较对相结合鉴定KISS1基因单核苷酸多态性(SNP)位点,分析SNP位点对柯乐猪繁殖指标的影响。结果显示在KISS1基因第1外显子处发现g.64835073 A>G同义突变,在第1内含子处发现g.64835529 G>A突变,2个SNP位点均产生3种基因型,均为中度多态位点,且均符合Hardy-Weinberg平衡;g.64835073 A>G位点对5个繁殖指标的影响未能达到显著水平(P>0.05);其中g.64835529 G>A位点对总产仔数、初生个体重、断奶个体重的影响达显著水平,揭示g.64835529 G>A位点可能作为猪产仔数的一个分子标记。
To understand the effect of KISS1 on reproductive traits of pig.Direct sequencing of PCR products was combined with artificial comparison to identify the SNPs of KISS1 gene in 78 Kele pigs and analyze the influence of SNP sites on reproductive indexes.The results showed that the g.64835073 AG and g.64835529 GA mutation were found in exon 1 and intron 1 of KISS1gene,respectively.The two SNPs sites produced three genotypes,all of which were moderately polymorphic sites and in line with Hardy-Weinberg equilibrium.The effects of g.64835073 AG on five reproductive indices were not significant(P0.05),the effects of G.64835529 G A locus on total litter size,birth weight and weaning weight were significant,and suggested that the g.64835529 GA site may be a molecular marker of pig litter size.
