新型光化纳米载药系统的构建及其对荷瘤鼠舌鳞癌模型治疗作用研究

摘要 目的 本研究合成了一种新型光化联合纳米载药系统,以期为舌鳞癌综合治疗提供新的策略。方法 通过水热合成法及盐酸蚀刻法,制备介孔普鲁士蓝纳米粒子(PBNP’s);用透明质酸-聚乙二醇共聚物(HA-PEG)进行修饰实现肿瘤主动靶向,形成PBNP’s-HA-PEG(简称PH);继而装载顺铂纳米颗粒,完成新型纳米给药体系Nano CDDP/PBNP’ s-HA-PEG(简称NCPH)的制备;随后对该体系粒径分布、靶向性、药物释放性能及光敏特性等进行表征;最后进行体内外抑瘤实验及生物安全性评价。结果 PH粒径在100 nm左右,靶向性良好,光热性能稳定,光照条件下可加快药物释放速度;体内外抑瘤试验结果表明NCPH在一定浓度范围内可以显著抑制体内外肿瘤生长;生物安全性评估发现,该载药系统毒副作用小,耐受性良好。结论 成功制备了新型载药系统NCPH,实现了抗舌鳞癌的光化联合治疗,生物安全性良好。 Objective In this study,we synthesized a new photochemical nano-drug delivery system to provide a new strategy for the comprehensive treatment of tongue squamous cell carcinoma.Methods Mesoporous Prussian blue nanoparticles(PBNP' s) were prepared by hydrothermal synthesis and hydrochloric acid etching;Then PBNP's were modified with hyaluronic acid-polyethylene glycol copolymer(HA-PEG) to achieve active tumor targeting and form PBNP's HA-PEG(PH);And then PBNP's HA-PEG were loaded with cisplatin nanoparticles to construct the new nano drug delivery system Nano CDDP/PBNP's HA-PEG(NCPH);Then the particle size distribution,targeting,drug release performance and photosensitivity of the system were tested;Finally,the antitumor experiments in vitro and in vivo and the biological safety evaluation were carried out.Results The particle size of PBNP's HA-PEG was about 100 nm,the targeting property was good,the photothermal property was stable,and the drug release rate could be accelerated under light conditions;The results of antitumor tests in vitro and in vivo showed that Nano CDDP/PBNP's HA-PEG could significantly inhibit tumor growth in vitro and in vivo within a certain concentration range;The biosafety evaluation found that the drug delivery system had few side effects and good compatibility.Conclusions A new drug delivery system Nano CDDP/PBNP's HA-PEG has been prepared successfully,and the combined photochemical treatment of tongue squamous cell carcinoma has been achieved with good biological safety.
