Tbx2和Tbx3在不同毛色绵羊皮肤中的表达与定位

The expression and localization of Tbx2 and Tbx3 in skin of sheep with different coat colors

摘要 为阐明Tbx2、Tbx3在绵羊毛色形成中的作用,本试验研究了T-box转录因子2(T-boxtranscription factor 2,Tbx2)、T-box转录因子3(T-box transcription factor 3,Tbx3)在不同毛色哈萨克绵羊皮肤中的表达与定位。选取黑色、白色12月龄雌性哈萨克绵羊各3只,取背部皮肤组织,通过qRT-PCR、Western-blot检测不同毛色皮肤中Tbx2和Tbx3 mRNA及蛋白相对表达量,免疫组化技术对Tbx2和Tbx3在绵羊皮肤组织中蛋白进行定位分析。结果,Tbx2基因在白色绵羊皮肤中mRNA相对表达量极显著高于黑色绵羊(P<0.01),白色绵羊的皮肤中Tbx2蛋白表达量显著高于黑色绵羊(P<0.05);Tbx3基因在黑色绵羊皮肤中mRNA及蛋白相对表达量均显著高于白色绵羊(P<0.05);Tbx2蛋白在白色绵羊毛囊组织的毛基质部表达,在黑色绵羊毛囊组织的外根鞘部表达。Tbx3蛋白在白色绵羊毛囊组织的外根鞘部和毛基质部表达,在黑色绵羊毛囊组织的外根鞘部表达。结果表明,Tbx2和Tbx3在不同毛色绵羊皮肤中表达不同,Tbx2和Tbx3可能通过诱导黑色素的生成进而调控哈萨克绵羊毛色生成过程。 In order to illustrate the role of Tbx2 and Tbx3 in the formation of sheep coat color,the expression and localization of T-box transcription factor 2(Tbx2)and T-box transcription factor 3(Tbx3)in the skin of sheep with different coat colors were studied in the experiment.Three female Kazakh sheep aged 12 months and three white female Kazakh sheep aged 12 months were selected in the ex-periment.Take the back skin tissues,and the relative expression levels of Tbx2 and Tbx3 mRNA and pro-tein in different skin colors were detected by qRT-PCR and Western-blot.The protein localization of Tbx2 and Tbx3 in sheep skin tissues was analyzed by immunohistochemistry.In result,the relative mRNA expression level of Tbx2 gene in white sheep was significantly higher than that in black sheep(P<0.01),and the protein expression level of Tbx2 in white sheep was significantly higher than that in black sheep(P<0.05).The mRNA and protein relative expression levels of Tbx3 gene in black sheep were significantly higher than those in white sheep(P<0.05).Tbx2 protein was expressed in the hair matrix of white sheep hair follicle tissue and in the outer root sheath of black sheep hair follicle tissue.Tbx3 protein was expressed in the outer root sheath and hair matrix of white sheep hair follicles,and in the outer root sheath of black sheep hair follicles.In conclusion,Tbx2 and Tbx3 are expressed differ-ently in skin of sheep with different hair colors,and Tbx2 and Tbx3 may regulate the hair color genera-tion process of Kazakh sheep by inducing melanin production.
