长链非编码RNA NKILA和微小RNA-485-5p在慢性鼻窦炎伴鼻息肉中的表达研究

摘要 目的:分析长链非编码RNA NKILA(LncRNA NKILA)和微小RNA-485-5p(miR-485-5p)在慢性鼻窦炎伴鼻息肉(CRSwNP)中的表达及意义。方法:选取慢性鼻窦炎(CRS)患者93例,分为伴鼻息肉(CRSwNP)组49例,不伴鼻息肉(CRSsNP)组44例,另选取单纯鼻中隔偏曲患者36例作为对照组。采用视觉量表评分(VAS)、Lund-Kennedy鼻内镜评分、Lund-Mackay鼻窦CT评分(CT评分)评估所有受试对象的疾病严重程度;取CRSwNP组患者息肉组织、CRSsNP组钩突组织及对照组下鼻甲组织,qRT-PCR法检测组织中NKILA和miR-485-5p水平;采用R语言limma包分析LncRNA差异表达;Pearson法分析各变量的相关性;无序多分类Logistics回归分析影响CRSwNP的因素;ROC曲线分析NKILA、miR-485-5p水平对CRSwNP的诊断价值。结果:CRSwNP组、CRSsNP组及对照组NKILA水平依次降低,miR-485-5p水平依次升高,两两比较差异有统计学意义(P<0.05)。CRSwNP组息肉组织中NKILA与miR-485-5p呈负相关(P<0.05)。GSE136825芯片差异分析结果显示,NKILA显著上调。无序多分类Logistics回归分析显示,随着NKILA水平升高,CRSwNP患病风险不断增大,miR-485-5p水平升高可降低CRSwNP患病风险。ROC曲线显示,NKILA、miR-485-5p均对CRSwNP有较高的诊断价值。结论:CRSwNP患者息肉组织中NKILA高表达,miR-485-5p低表达,二者均与患者疾病严重程度有关,且对CRSwNP有一定的诊断价值。 Objective:To analyze the expression and significance of long non-coding RNA NKILA(LncRNA NKILA)and microRNA-485-5p(miR-485-5p)in chronic rhinosinusitis with nasal polyps(CRSwNP).Methods:A total of 93 patients with chronic rhinosinusitis(CRS)were selected and divided into CRSwNP group(49 cases)and without nasal polyps(CRSsNP)group(44 cases).In addition,36 patients with simple septal deviation were selected as the control group.Disease severity was assessed in all subjects using the Visual Scale Score(VAS),the Lund-Kennedy Endoscopy Score,and the Lund-Mackay Sinus CT Score(CT Score).Polyp tissue of patients in CRSwNP group,uncinate process tissue of CRSsNP group and inferior turbinate tissue of control group were collected,and the levels of NKILA and miR-485-5p in the tissues were detected by qRT-PCR.Differential LncRNA expression was analyzed by R language limma package.Pearson method was used to analyze the correlation of variables.Unordered multi-classification Logistics regression analyze the factors that affecting CRSwNP.ROC curve was used to analyze NKILA,miR-485-5p levels for the diagnostic value of CRSwNP.Results:The levels of NKILA decreased in CRSwNP group,CRSsNP group and control group,and the level of miR-485-5p increased sequentially,and the difference between the two was statistically significant(P<0.05).In CRSwNP group,NKILA in polyp tissue was negatively correlated with miR-485-5p(P<0.05).GEO database analysis and mapping of head and neck squamous cell carcinoma volcanoes revealed that NKILA was significantly up-regulated.Unordered multi-classification Logistics regression analysis revealed that the risk of CRSwNP increased as the NKILA levels increased and increased mir-485-5p levels can decrease the risk of CRSwNP.ROC curve showed that NKILA and miR-485-5p had high diagnostic value for CRSwNP.Conclusion:The polyps of CRSwNP patients have high expression of NKILA and low expression of miR-485-5p,both of which are related to the severity of the disease,and have certain diagnostic value for CRSwNP.
