香合欢种子萌发及不定芽诱导研究 被引量:2

摘要 为筛选香合欢(Albizia odoratissima)破除种子硬实和不定芽诱导的有效方法,使其在短期内大量萌发和快速扩繁,以香合欢种子为材料,研究不同水温和浓硫酸处理对种子萌发的影响,选出最佳的硬实破除方式;采用不同灭菌方式,选出适合种子萌发的灭菌溶液及浓度;以无菌苗的茎段作为外植体,探究不同激素浓度对茎段初代培养及增殖培养的影响。结果表明,80℃热水对破除种子硬实的效果较好,其发芽率为86.67%。8%H2O2灭菌方式最适合香合欢苗生长,其苗高、总根长分别为13.0和32.5 cm。1/2MS+6-BA 1.5 mg/L+IBA 0.2 mg/L为最佳初代培养基,其增殖系数为1.77;3/4MS+6-BA1.5 mg/L+NAA 0.4 mg/L+TDZ 0.15 mg/L为最佳增殖培养基,其增殖系数为4.67。 In order to screen effective methods for breaking hardseededness and adventitious bud induction of Albizia odoratissima seeds for mass germination and rapid expansion in a short period,taking A.odoratissima seeds as materials,effects of different water temperatures and concentrated sulfuric acid treatments on seed germination were analyzed and the best breaking hardseededness method was selected.Suitable sterile solution and concentration for seed germination were selected by treating seeds with different sterilization methods.Stem segments from sterile seedlings were used as explants to explore effects of different hormone concentrations on primary culture and proliferation culture of stem segments.Results showed that 80℃hot water had better effect on breaking hardseededness,with germination rate of 86.67%.The most suitable method for seed germination and growth of A.odoratissima seedlings was 8%H2O2 sterilization method,with seedling height and total root length of 13.0 and 32.5 cm,respectively.Optimal initial culture medium was 1/2MS+6-BA 1.5 mg/L+IBA 0.2 mg/L,with proliferation coefficient of 1.77.Optimal proliferation medium was 3/4MS+6-BA 1.5 mg/L+NAA 0.4 mg/L+TDZ 0.15 mg/L,with proliferation coefficient of 4.67.
