miR-599靶向YAP1对卵巢癌细胞迁移及侵袭的影响

Effects of miR⁃599 targeting YAP1 on the migration and invasion of ovarian cancer cells

目的:探讨miR-599靶向Yes相关蛋白1(YAP1)对卵巢癌细胞迁移及侵袭的影响。方法:采用qRT-PCR检测正常卵巢上皮细胞IOSE-80与卵巢癌细胞SKOV3中miR-599的表达;将SKOV3细胞分为6组:空白组、miR-NC组、miR-599 mimics组、siRNA-NC组、YAP1 siRNA组、YAP1 siRNA+miR-599 inhibitor组,划痕实验检测各组细胞迁移;Transwell实验检测各组细胞侵袭;Western Blot检测各组细胞中YAP1蛋白表达;双萤光素酶实验验证miR-599与YAP1的关系。结果:miR-599在SKOV3细胞中的表达水平显著低于IOSE-80细胞(P<0.05);过表达miR-599或沉默YAP1均可抑制SKOV3细胞的迁移和侵袭;miR-599可靶向调控YAP1表达;抑制miR-599可逆转沉默YAP1对SKOV3细胞迁移和侵袭的抑制作用。结论:过表达miR-599通过下调YAP1来抑制SKOV3细胞的迁移和侵袭。

Objective:To investigate the effects of miR⁃599 targeting Yes⁃associated protein 1(YAP1)on the migration and invasion of ovarian cancer cells.Methods::qRT⁃PCR was used to detect the expres⁃sion of miR⁃599 in normal ovarian epithelial cells IOSE⁃80 and ovarian cancer cells SKOV3.SKOV3 cells were divided into six groups as blank group,miR⁃NC group,miR⁃599 mimics group,siRNA⁃NC group,YAP1 siRNA group,and YAP1 siRNA+miR⁃599 inhibitor group.Scratch test was adopted to detect cell migration in each group.Transwell test was adopted to detect cell invasion in each group.Western blot was adopted to detect YAP1 protein expression in each group of cells.The dual luciferase experiment was adopted to verify the relationship between miR⁃599 and YAP1.Results::The expression level of miR⁃599 in SKOV3 cells was significantly lower than that of IOSE⁃80 cells(P<0.05);Overexpression of miR⁃599 or silencing YAP1 could inhibit the migration and invasion of SKOV3 cells;miR⁃599 could target and regulate YAP1 expression;Inhibition of miR⁃599 could reverse the inhibitory effect of silencing YAP1 on SKOV3 cell migration and invasion.Conclusion::Overexpression of miR⁃599 inhibits the migration and invasion of SKOV3 cells by down⁃regulating YAP1.
