摘要
目的:探讨安妥沙星体外抗结核活性,并与莫西沙星(moxifloxacin,MXF)、左氧氟沙星(levofloxacin,LVFX)比较,明确是否存在抗结核优势。方法:结核分枝杆菌(mycobacterium tuberculosis,M.tuberculosis)标准菌株为H37Ra,30例对利福平(rifampicin,RFP)敏感结核分枝杆菌菌株和16例对RFP耐药的结核分枝杆菌菌株来自于某市传染病医院细菌室的临床分离株。采用7H9+OADC/ADC液体培养基,使用不同浓度的试验药物进行抑菌试验,判读MIC结果,H37Ra孔板行酶标仪监测96孔板OD600值。结果:H37Ra试验药物孔板对应的OD600值随药物浓度增加而下降,安妥沙星和MXF的MIC均为0.5μg/mL,LVFX的MIC是1μg/mL。对RFP敏感的结核分枝杆菌株组的MIC90,同H37Ra试验结果。三种化合物对RFP耐药的结核分枝杆菌株组的MIC90分别是4μg/mL,1μg/mL和>8μg/mL。结论:安妥沙星体外存在抗结核活性,对RFP敏感的结核分枝杆菌抗结核疗效与MXF相同,而对RFP耐药的结核分枝杆菌抗结核疗效不及莫西沙星,但优于LVFX。
Objective To explore the anti-tuberculosis activity of antofloxacin in vitro,and compare it with moxifloxacin(MXF)and levofloxacin(LVFX)to determine whether there is an anti-tuberculosis advantage.Methods The standard strain of Mycobacterium tuberculosis(M.tuberculosis)was H37Ra.30 strains of M.tuberculosis sensitive to rifampicin(RFP)and 16 strains of M.tuberculosis resistant to RFP were clinically isolated from the bacterial room of a municipal infectious disease hospital.Using H9+OADC/ADC liquid culture medium,antibacterial tests were conducted with different concentrations of experimental drugs.MIC results were interpreted and the OD600 value of the 96-well plate was monitored by a microplate reader on the H37Ra plate.Results The OD600 value corresponding to the orifice plate of H37Ra test drug decreased with the increase of drug concentration.The MIC of antofloxacin and MXF was both 0.5μg/mL,and the MIC of LVFX was 1μg/mL.MIC90 of M.tuberculosis strain group sensitive to RFP was the same as H37Ra test result.The MIC90 of the three compounds against the RFP-resistant M.tuberculosis strain was 4μg/mL,1μg/mL and>8μg/mL.Conclusion Antofloxacin has anti-tuberculosis activity in vitro.The anti-tuberculosis effect on RFP-sensitive M.tuberculosis is the same as that of MXF,while the anti-tuberculosis effect on RFP-resistant M.tuberculosis is not good as moxifloxacin,but better than LVFX.
