丹桃养肝丸对四氯化碳致肝纤维化大鼠的保护作用研究

Protective effect of Dantao Yanggan pill on carbon tetrachloride induced liver fibrosis in rats

目的观察丹桃养肝丸对四氯化碳(CCl 4)诱导肝纤维化大鼠的保护作用。方法取80只SD大鼠,随机选择14只作为对照组,其余大鼠采用连续9周轮替四肢内侧皮下注射CCl 4方法制备肝纤维化模型。将造模成功大鼠随机分成5组,秋水仙碱组给予秋水仙碱0.75 mg/kg灌胃,丹桃养肝丸高、中、低剂量组分别给予6 g/kg、3 g/kg、1.5 g/kg丹桃养肝丸灌胃,对照组和模型组给予去离子水灌胃,均1次/d,连续灌胃21 d。大鼠处死后,全自动生化分析仪检测血清中丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、碱性磷酸酶(ALP)、γ-谷氨酰转移酶(GTT)水平,计算AST/ALT值;分离肝、脾和胸腺,计算脏器指数;ELISA法检测肝组织中羟脯氨酸(Hyp)含量;HE染色观察肝脏组织纤维化程度,并根据评分标准进行评分。结果与对照组相比,模型组血清ALT、AST、GGT、ALP水平有上升趋势,AST/ALT有下降趋势,肝组织中Hyp含量、肝纤维化评分均明显升高(P均<0.05)。与模型组比较,丹桃养肝低剂量组血清AST水平,丹桃养肝中、低剂量组AST/ALT值,秋水仙碱组和丹桃养肝高、中剂量组肝组织中Hyp含量和肝纤维化评分均明显降低(P均<0.05),秋水仙碱组和丹桃养肝高、中剂量组胸腺指数均明显增高(P均<0.05)。结论丹桃养肝丸能够改善肝纤维化大鼠肝功能,增强脏器免疫功能,减少胶原纤维生成,减轻肝纤维化损伤。

Objective It is to explore the protective effect of Dantao Yanggan pill on liver fibrosis induced by carbon tetrachloride(CCl 4)in rats.Methods Eighty SD rats were selected,in which 14 ones were randomly selected as control group,and the rest rats were given CCl 4 by subcutaneous injection into their inner extremities in succession for 9 weeks to establish models of liver fibrosis.The successfully modeled rats were randomly divided into 5 groups:the colchicine group was given colchicine 0.75 mg/kg by gavage,the high,medium and low dose groups of Dantao Yanggan pill were given Dantao Yanggan pills 6 g/kg,3 g/kg and 1.5 g/kg by gavage,respectively,and the control group and model group were given deionized water by gavage,all once daily,continuously treated for 21 days.The control group and the model group were given deionized water and administered by gavage continuously for 21 days.After the rats were executed,the levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase(ALP),andγ-glutamyltransferase(GTT)in serum were detected by automatic biochemical analyzers,and the values of AST/ALT were calculated;the livers,spleens,and thymus glands were isolated,and the index of the organs was calculated;the content of hydroxyproline(Hyp)in liver tissue was detected by ELISA;the degree of fibrosis in liver tissue was observed by HE staining,and was scored according to scoring criteria.Results Compared with the control group,the levels of ALT,AST,GGT,and ALP in serum in the model group showed an upward trend,while AST/ALT showed a downward trend,the content of Hyp in liver tissue and liver fibrosis score were significantly higher than those in the control group(both P<0.05).Compared with the model group,the level of AST in serum in the low dose group of Dantao Yanggan pill,the AST/ALT values in the medium and low dose groups of Dantao Yanggan pill,the content of Hyp and liver fibrosis score in liver tissue in the colchicine group and the high and medium dose groups of Dantao Yanggan pill were significantly reduced(all P<0.05),and the thymus index of the colchicine group and the high and medium dose groups of Dantao Yanggan pill were significantly increased(all P<0.05).Conclusion Dantao Yanggan pill can improve liver function,improve organ immune function,reduce collagen fiber production,and alleviate hepatic fibrosis injury in rats with hepatic fibrosis.
