摘要
目的:建立甘草多糖胶囊中甘草多糖含量测定的方法。方法:以苯酚和硫酸的加入量、反应时间等为考察因素,采用正交实验设计优化苯酚-硫酸法测定甘草多糖含量的显色条件。并采用紫外分光光度法,以葡萄糖为对照品,于波长490 nm处测定甘草多糖吸光度,计算总多糖的含量。结果:甘草总多糖含量测定的最佳显色条件为苯酚用量1 mL,硫酸用量5 mL,放置时间30 min。甘草多糖在0.0621~0.1656 mg·mL^(-1)浓度范围内线性关系良好,回归方程为y=0.0048x+0.0252,r=0.9995;精密度、稳定性、重现性良好;平均加样回收率为99.5%,RSD=3.0%(n=9)。结论:该方法简便、准确、重复性好,可用于甘草多糖胶囊中总多糖的含量测定。
Objective To establish a method for the determination of total polysaccharides in Glycyrrhiza polysaccharide capsules.Methods Taking the amount of phenol and sulfuric acid,reaction time and so on as the investigation factors,the orthogonal experimental design was adopted to optimize the color development conditions for the determination of Glycyrrhiza polysaccharide content by phenol-sulfuric acid method.The absorbance of Glycyrrhiza polysaccharide was measured at 490nm by ultraviolet spectrophotometry,and the content of total polysaccharide was calculated.Results The best chromogenic conditions for the determination of total polysaccharide content in Glycyrrhiza uralensis Fisch were as follows:the dosage of phenol was 1 mL,the dosage of sulfuric acid was 5 mL,and the storage time was 30 min.The linear relationship of Glycyrrhiza polysaccharide is good in the concentration range of 0.0621~0.1656 mg·mL^(-1),and the regression equation is y=0.0048x+0.0252,r=0.9995;Good precision,stability and reproducibility;The average recovery rate is 99.5%,RSD=3.0%(n=9).Conclusion The method is simple,accurate and reproducible,and can be used for the determination of total polysaccharides in Glycyrrhiza polysaccharide capsules.
