外泌体miR-29对上皮性卵巢癌细胞的迁移、侵袭及血管生成能力的影响

Effects of exosomal miR-29 on migration,invasion and angiogenesis of epithelial ovarian cancer cells

摘要 目的探究外泌体微小RNA29(miR-29)对上皮性卵巢癌(EOC)细胞的迁移、侵袭及血管生成能力的影响,及其作用机制。方法体外培养EOC细胞系SK-OV-3细胞,并分为空白组(不做任何处理)、对照组(加入空白病毒外泌体)和实验组(加入miR-29高表达的病毒外泌体)。用实时荧光定量聚合酶链反应检测细胞miR-29的表达情况,分别用CCK-8、划痕实验及Transwell实验检测各组细胞的增殖、迁移及侵袭能力,用小管生成实验检测细胞血管生成能力。结果空白组、对照组和实验组的miR-29相对表达量分别为0.52±0.06、0.54±0.05和1.35±0.15,小管形成数分别为(11.18±1.20)、(11.26±1.25)和(5.18±0.61)个,细胞增殖抑制率分别为0.00、(0.05±0.01)%和(12.75±0.15)%,划痕愈合率分别为(26.08±2.94)%、(25.46±3.06)%和(18.81±1.86)%,细胞侵袭细胞数分别为(96.18±9.81)、(95.61±9.98)和(42.38±4.36)个。空白组的上述指标与对照组比较,差异均无统计学意义(均P>0.05);实验组的上述指标与空白组和对照组比较,差异均有统计学意义(均P<0.05)。结论外泌体miR-29的高度表达能够抑制EOC细胞增殖、迁移及侵袭能力,降低血管生成能力。 Objective To investigate the effect and mechanism of exosomal microRNA29(miR-29)on the migration,invasion and angiogenesis of epithelial ovarian cancer(EOC)cells.Methods The EOC line SK-OV-3 cells were cultured in vitro and divided into blank group(without any treatment),control group(adding blank virus exosomes)and experimental group(adding virus exosomes with high expression of miR-29).The expression levels of miR-29 was detected by real-time polymerase chain reaction.The proliferation,migration,invasion abilities and angiogenesis abilities in each group were detected by CCK-8,scratch assay,Transwell assay and tubule formation assay.Results The relative expression levels of miR-29 in blank,control and experimental groups were 0.52±0.06,0.54±0.05 and 1.35±0.15;the number of SK-OV-3 tubule formation was 11.18±1.20,11.26±1.25 and 5.18±0.61;the inhibition rates of SK-OV-3 cell proliferation were 0.00,(0.05±0.01)%and(12.75±0.15)%;the scratch healing rates were(26.08±2.94)%,(25.46±3.06)%and(18.81±1.86)%;the number of invasive cells was 96.18±9.81,95.61±9.98 and 42.38±4.36,respectively.There were no significant differences in the above indexes between blank group and control group(all P<0.05).Compared with blank group and control group,the above indexes of experimental group were statistically significant(all P<0.05).Conclusion High expression of exosome miR-29 can inhibit EOC cells proliferation,migration and invasion,and reduce angiogenesis.
