摘要
为了探究噬菌体使用前后驴肠道微生物多样性及结构组成的变化,并借以评价噬菌体(vB-SabS-Sds2)的治疗效果。对80头驴的粪便使用沙门菌特异性引物invA进行聚合酶链式反应(polymerase chain reaction,PCR)检测,选择13头沙门菌检测阳性驴,将13头阳性驴在同等条件单独隔离饲养,在饲喂噬菌体前后掏肛采集粪便,利用16S rRNA高通量测序技术,对两组驴粪便在微生物多样性方面(结构和组成)存在的差异进行比较。结果表明:与噬菌体饲喂前相比,饲喂后的驴肠道菌群丰富度显著升高(P<0.05),目标宿主肠杆菌科细菌数量显著降低,且部分益生菌相对丰度升高;含多种临床条件致病菌的假单胞菌属和梭状芽胞杆菌属细菌的相对丰度显著降低。因此,饲喂噬菌体不会对驴肠道正常菌群的多样性造成影响,而且能改善驴肠道健康状况和微生态环境。
Previous studies have shown that antibiotics will affect the microbial diversity of donkey intestinal tract,but the bactericidal effect of bacteriophage is specific,and the bacteriophage of pathogenic bacteria does not destroy the normal intestinal flora.The purpose of this study was to explore the changes of intestinal flora diversity and structural composition of donkey before and after phage use,and to evaluate the therapeutic effect of phage(vB-SabS-Sds2).In this experiment,the feces of 80 donkeys were detected by PCR with Salmonella specific primer invA,and then 13 donkeys with Salmonella positive were isolated and raised under the same conditions.The feces of 13 Salmonella positive donkeys were collected from the anus before feeding the phage,and the feces were also collected from the anus after feeding the phage for three days.The differences in microbial diversity(structure and composition)between the two groups of donkey feces were compared by 16S rRNA high-throughput sequencing technology.The results showed that the intestinal microflora richness of donkey fed with phage was significantly higher than that before phage feeding(P<0.05).After phage feeding,the number of Enterobacteriaceae bacteria in the target host decreased significantly,and the relative abundance of some probiotics increased;The relative abundance of Pseudomonas and Clostridium containing a variety of clinical condition pathogenic bacteria also decreased significantly.In conclusion,phage application will not affect the normal flora diversity of donkey intestine,and can even improve donkey intestinal health and micro ecological environment.
