lncRNA LINC00460在结直肠癌组织中的表达及其对细胞增殖、侵袭和迁移能力的影响

The expression of lncRNA LINC00460 in colorectal cancer tissues and its impact on cell proliferation,invasion and migration ability

目的:探讨长链非编码RNA(lncRNA)LINC00460在结直肠癌组织中的表达及对结直肠癌细胞增殖、侵袭和迁移的影响。方法:GEPIA在线数据库和实时荧光定量PCR(qPCR)分析检测lncRNA LINC00460在结直肠癌组织、结直肠癌细胞株(HCT-8、HCT-116、SW480、SW620)及结直肠黏膜上皮细胞NCM460中的表达,进一步分析LINC00460表达与结直肠癌患者临床病理特征的关系。将LINC00460过表达质粒及阴性对照(pcDNA3.1-LINC00460和pcDNA3.1-NC)转染HCT-8细胞,将LINC00460小干扰RNA及阴性对照(si-LINC00460和si-NC)转染SW620细胞,分别采用CCK-8实验、克隆形成实验和Transwell实验检测细胞增殖能力和侵袭迁移能力,采用Western blot实验检测cyclin D1、p21、E-cadherin、N-cadherin和Vimentin蛋白的表达。结果:与癌旁组织和NCM460细胞相比较,LINC00460在结直肠癌组织及结直肠癌细胞株(HCT-8、HCT-116、SW480、SW620)中高表达(均P<0.05),并与肿瘤分化程度、患者TNM分期、淋巴结转移和Ki-67增殖指数呈正相关(均P<0.05);下调LINC00460表达引起p21和E-cadherin表达增加,cyclin D1、N-cadherin和Vimentin表达降低,抑制SW620细胞的增殖、侵袭和迁移,上调LINC00460表达引起p21和E-cadherin表达降低,cyclin D1、N-cadherin和Vimentin表达增加,增强HCT-8细胞的增殖、侵袭和迁移。结论:lncRNA LINC00460在结直肠癌组织及细胞中高表达,并与结直肠癌患者临床病理特征相关,沉默或过表达LINC00460可减弱或增强结直肠癌细胞增殖、侵袭及迁移能力。

Objective:To investigate the expression of long non-coding RNA(lncRNA)LINC00460 in colorectal cancer(CRC)tissues,and its effects on the proliferation,invasion and migration of CRC cells.Methods:The expression of lncRNA LINC00460 in CRC tissues,CRC cell lines(HCT-8,HCT-116,SW480,SW620)and colorectal mucosal epithelial cells NCM460 was analyzed by using the Gene Expression Profiling Interactive Analysis(GEPIA)database and quantitative polymerase chain reaction(qPCR).The correlation between the expression level of LINC00460 and clinicopathological parameters of CRC patients was further analyzed.After HCT-8 and SW620 cells were transfected with pcDNA3.1-LINC00460,pcDNA3.1-NC,si-LINC00460 or si-NC,the proliferation,invasion and migration of CRC cell lines(HCT-8 and SW620)was detected by CCK-8 assay,clonal formation assay,and Transwell assay,respectively.And the Western blot assay was used to detect the expressions of cyclin D1,p21,E-cadherin,N-cadherin and Vimentin.Results:Compared with para-carcinoma tissues and NCM460 cells,lncRNA LINC00460 was highly expressed in CRC tissues and CRC cell lines(HCT-8,HCT-116,SW480,SW620)(P<0.05),and the high level of LINC00460 was significantly correlated with tumor differentiation,TNM stage,lymph node metastasis and Ki-67 expression(all P<0.05).Down-regulated the expression of lncRNA LINC00460 can increase the expression of p21 and E-cadherin,decrease the expression of cyclin D1,N-cadherin and Vimentin,and inhibit the proliferation,invasion and metastasis of SW620 cells.Up-regulated expression of lncRNA LINC00460 can decrease the expression of p21 and E-cadherin,increase the expression of cyclin D1,N-cadherin and Vimentin,and promoted the proliferation,invasion and metastasis of HCT-8 cells.Conclusion:lncRNA LINC00460 is highly expressed in CRC tissues and cells,and its expression is significantly correlated to the clinical parameters of CRC patients.Up-regulated or down-regulated the expression of LINC00460 could correspondingly promote or inhibit the proliferation,invasion and migration of CRC cells.
