HPLC法研究不同产地蔓荆子果实及花萼中蔓荆子黄素含量的差异

摘要 目的收集不同产地的蔓荆子果实及花萼并测定其蔓荆子黄素的含量,探讨不同产地蔓荆子的品质及去除花萼的影响。方法用高效液相色谱法分别对蔓荆子果实及花萼中蔓荆子黄素含量进行测定。色谱柱为Agilent ZORBAX SB-C_(18)(250 mm×4.6 mm,5μm);流动相为甲醇4 mL·L^(-1)磷酸溶液(60∶40);检测波长为258 nm;柱温为30℃;流速为1.0 mL·min^(-1)。结果蔓荆子中蔓荆子黄素的回归方程为y=13001 x-3.59(r=0.9996),线性范围为14.5~72.5μg·mL^(-1)。结论不同产地的蔓荆子药材品质差异较大,其蔓荆子黄素含量不同。同时花萼中同样含有蔓荆子黄素,且部分地区所产的蔓荆子花萼中蔓荆子黄素的含量较高,去除花萼将影响药材品质。 Objective To analyze the content of vitexiarpin in fruits and calyx of Viticis fructus from different producing areas,so as to study the quality of Viticis fructus from different producing areas and the effect of clayx removal.Methods High performance liquid chromatography(HPLC)was used to analyze the content of vitexiarpin in fruits and calyx of Viticis fructus.Agilent ZORBAX SB-C_(18)(250 mm×4.6 mm,5μm)was used with methanol-4 mL·L^(-1) phosphoric acid(60∶40)as the mobile phase.The detection wavelength was 258 nm,the column temperature was 30℃,and the flow rate was 1.0 mL·min^(-1).Results The regression equation was y=13001 x-3.59(r=0.9996),and the linear range was 14.5-72.5μg·mL^(-1).Conclusion There were obvious differences in the content of vitexiarpin in Viticis fructus from different producing areas.The vitexiarpin also existed in calyx,and the content was higher than in fruits in some areas.Therefore the removal of calyx affects the quality of Viticis fructus.
