女贞子酒蒸前后分别配方的二至丸HPLC指纹图谱差异分析

Difference Analysis of HPLC fingerprint of Erzhi Pill formulated of Ligustri Lucidi Fruc⁃tus before and after steaming with wine

摘要 目的比较女贞子酒蒸前后分别配方的二至丸HPLC指纹图谱差异,研究女贞子酒蒸对二至丸化学成分的影响。方法采用HPLC法测定不同批次的女贞子生品与酒蒸品分别配方的二至丸指纹图谱,并进行相似度评价;采用主成分分析(PCA)、正交偏最小二乘判别分析(OPLS-DA)法研究女贞子生品与酒蒸品分别配方的二至丸HPLC指纹图谱差异,筛选出其差异成分。结果女贞子酒蒸前后分别配方的二至丸HPLC指纹图谱具有35个共有峰,10批生女贞子配方的二至丸HPLC指纹图谱相似度为0.936~0.994,10批酒蒸女贞子配方的二至丸HPLC指纹图谱相似度为0.937~0.995;女贞子酒蒸前后配方的二至丸HPLC指纹图谱具有显著差异,共筛选出羟基酪醇、特女贞苷、女贞苷G13、女贞酸、红景天苷、毛蕊花糖苷、酪醇、油女贞苷、松果菊苷等14种差异成分。结论女贞子酒蒸对二至丸的化学成分影响显著,建立的指纹图谱结合PCA、OPLS-DA可有效区分女贞子生品与酒蒸品配方的二至丸,辨识出的差异成分为二至丸质量标志物的选择提供了科学依据。 Objective The differences of HPLC fingerprints of Erzhi pills formulated of Ligustri Lucidi Fructus before and after steaming with wine were compared to study the effect of wine-steamed Ligustri Lucidi Fructus on the chemical constituents of Erzhi pills.Methods The fingerprints of Erzhi pills of different batches of raw and wine-steamed Ligustri Lucidi Fructus were de⁃termined by HPLC,and the similarity was evaluated,and the differences of HPLC fingerprints of raw and wine-steamed Ligustri Lucidi Fructus were studied by principal component analysis(PCA)and orthogonal partial least square discriminant analysis(OPLS-DA).Results The HPLC fingerprints of Erzhi pills formulated of OPLS-DA before and after steaming with wine have 35 common peaks.The HPLC fingerprint similarity of 10 batches of Erzhi pills formulated of raw Ligustri Lucidi Fructus is 0.936~0.994.The HPLC fingerprint similarity of 10 batches of Erzhi pills formulated of wine-steamed Ligustri Lucidi Fructus is 0.937~0.995.There were significant differences in HPLC fingerprints of Erzhi pills formulated of Ligustri Lucidi Fructus before and after steaming with wine.A total of 14 differential components were screened,such as hydroxytyrosol,specnuezhenide,nuezhe⁃noside G13,nuezhenidic acid,salidroside,verbascoside,tyrosol,oleonuezhenide,echinoside and so on.Conclusion Wine-steamed of Ligustri Lucidi Fructus has a significant effect on the chemical composition of Erzhi pills.The established fingerprint combined with PCA and OPLS-DA can effectively distinguish Erzhi pills formulated of raw products and wine-steamed products of Ligustri Lucidi Fructus.The identified different components provide a scientific basis for the selection of quality markers of Erzhi pills.
