经典名方一贯煎颗粒HPLC指纹图谱及化学模式识别研究

Study on HPLC Fingerprint and Chemical Pattern Recognition of the Classical Famous Prescription Yiguan Decoction Granules

摘要 目的 建立一贯煎颗粒HPLC指纹图谱,为该经典名方颗粒制剂的开发以及质量控制提供参考。方法 建立15批一贯煎颗粒的HPLC指纹图谱,应用“中药色谱指纹图谱相似度评价系统”软件(2012版)进行相似度分析,并对共有峰进行归属。运用聚类分析(HCA)、主成分分析(PCA)、正交偏最小二判别(OPLS-DA)对一贯煎颗粒进行质量分析。结果 15批一贯煎颗粒的相似度均大于0.951,共标定25个共有峰,指认出12、17、20、22、23号色谱峰,分别为香草酸、阿魏酸、毛蕊花糖苷、异类叶升麻苷、补骨脂素;其中1、3号峰归属于川楝子、生地黄、枸杞子,2、8、9、11、15、16号峰归属于枸杞子,4、6、21号峰归属于生地黄、枸杞子,5号峰归属于枸杞子、生地黄、川楝子,7、17、23号峰归属于当归、枸杞子,10号峰归属于枸杞子、当归、沙参,12、14号峰归属于枸杞子、川楝子、当归,13号峰归属于枸杞子、川楝子,18、19、20、22、24号峰归属于生地黄。结论 建立的一贯煎颗粒HPLC指纹图谱方法专属性强、分离度好、灵敏度高,可为经典名方一贯煎颗粒制剂的药效物质基础研究以及质量控制提供参考。 Objective To establish HPLC fingerprint of Yiguan decoction granules,providing reference for the development and quality control of the classic famous prescription granules.Methods The fingerprints of 15 batches of Yiguan decoction granules were established by HPLC gradient elution,and the similarity of the fingerprints was analyzed with Similarity Evaluation System for Chromatographic Fingerprint of Chinese Materia(2012)and assigned common peaks.The fingerprints data were evaluated by the hierarchical clustering analysis(HCA),principal component analysis(PCA)and orthogonal partial least squares-discrimi⁃nant analysis(OPLS-DA).Results The similarity of the 15 batches was bigger than 0.951.A total of 25 common peaks were calibrated,the chromatographic peaks 12,17,20,22 and 23 were identified as vanillic acid,ferulic acid,mullein glycoside,heterologous cimicifugin and psoralen respectively;Peaks 1 and 3 are attributed to Fructus Toosendan,Rehmannia glutinosa and the fruit of Chinese wolfberry,peaks 2,8,9,11,15 and 16 are attributed to the fruit of Chinese wolfberry,peaks 4,6 and 21 are attributed to Rehmannia glutinosa and the fruit of Chinese wolfberry,peak 5 is attributed to the fruit of Chinese wolfberry,Re⁃hmannia glutinosa and Fructus Toosendan,peaks 7,17 and 23 are attributed to Chinses angelica and the fruit of Chinese wolfber⁃ry,peak 10 is attributed to the fruit of Chinese wolfberry,Chinese angelica and Adenophora stricta,peaks 12 and 14 are attribu⁃ted to the fruit of Chinese wolfberry,Fructus Toosendan and Chinese angelica,peak 13 is attributed to the fruit of Chinese wolf⁃berry and Fructus Toosendan,peaks 18,19,20,22 and 24 are attributed to the Rehmannia glutinosa.Finally,ferulic acid is se⁃lected as the marker of its quality difference.Conclusion The established HPLC fingerprint method of Yiguan decoction granules has strong specificity,good resolution,and high sensitivity.It can provide a reference for the pharmacodynamic material basis and quality control of the classic famous prescription Yiguan decoction granules.
