蛤蚧真伪鉴定试剂盒的研制与性能评价

摘要 目的 蛤蚧作为我国的名贵中药材,资源少,市场需求量大,导致市场常有混伪品出现,因此研制蛤蚧真伪鉴定试剂盒,为蛤蚧药材鉴定提供新的方法。方法 以蛤蚧细胞色素C氧化酶亚基Ⅰ(CoⅠ)为靶基因,设计特异性引物,确定合适的PCR反应条件,研制蛤蚧真伪鉴定试剂盒。结果 扩增产物经过琼脂糖凝胶电泳后,正品可以扩增出约250~500bp之间(473bp)的扩增条带,而伪品无蹼壁虎和石龙子均没有条带。结论 研制出的蛤蚧真伪鉴定试剂盒特异性强,灵敏度高,重复性好,适用范围广,操作简单,结果准确,可以为蛤蚧的DNA分子鉴别提供技术保障,可以保证药源无误,药效可靠。 Objective Gecko is a rare and precious Chinese herbal medicine in China,with few resources and large market de⁃mand,resulting in the occurrence of mixed and counterfeit products in the market.Therefore,the development of gecko authentic⁃ity identification kit provides a new method for the identification of gecko.Methods Using CoⅠas the target gene,specific prim⁃ers were designed to determine the appropriate PCR reaction conditions and develop the gecko authenticity identification kit.Re⁃sults After agarose gel electrophoresis,the amplification bands between 250-500bp(473bp)could be obtained from the au⁃thentic products,,and the fake Gekko swinhonis and the fake Eumeces have no bands.Conclusion The developed gecko true and false identification kit has strong specificity,high sensitivity,good repeatability,wide application range,simple operation and ac⁃curate results,which can provide technical support for DNA molecular identification of gecko,guarantee the correct drug source and reliable efficacy.
