基于药效学结合响应面法优选穿王消炎片提取工艺研究

Optimization of Extraction Process of Chuanwang Xiaoyan Tablets(穿王消炎片)Based on Pharmacodynamics Combined with Response Surface Method

摘要 目的:优化穿王消炎片提取工艺,结合药效学研究对提取工艺进行验证,为工业生产提供指导。方法:使用人工汗液乙醇溶液作为提取液制备穿王干膏;单因素试验中以足肿胀抑制率为指标考察适宜提取温度,以浸膏收率为指标考察适宜人工汗液乙醇溶液用量,以足肿胀抑制率和AST值为指标考察适宜人工汗液乙醇浓度;采用Box-Behnken试验设计,以大鼠足肿胀抑制率、AST值、浸膏收率为综合评价指标,优化穿王消炎片提取工艺。结果:穿王消炎片最佳提取工艺:提取温度为63℃,人工汗液乙醇溶液用量为8.5倍量,人工汗液乙醇溶液浓度为27%。在此条件下综合评分值为89.12,理论值为89.24,与理论值相对误差小于5%。结论:本试验采用的方法和指标适合同时提取穿心莲、了哥王,所得提取物抗炎药效高、毒性低、副作用小。 Objective:To optimize the extraction process of Chuanwang Xiaoyan Tablets,and to verify the extraction process in combination with pharmacodynamic studies,so as to provide guidance for industrial production.Methods:The artificial sweat ethanol solution was used as the extract to prepare Chuanwang dry paste.In the single factor experiment,the suitable extraction temperature was investigated with the inhibition rate of foot swelling as the index,the suitable amount of ethanol solution of artificial sweat was investigated with the yield of extract as the index,and the suitable ethanol concentration of artificial sweat was investigated with the inhibition rate of foot swelling and AST value as the index.The Box-Behnken experimental design was used to optimize the extraction process of Chuanwang Xiaoyan Tablets with the inhibition rate of rat foot swelling,AST value and extract yield as comprehensive evaluation indexes.Results:The optimum extraction process of Chuanwang Xiaoyan Tablets was as follows:the extraction temperature was 63℃,the amount of artificial sweat ethanol solution was 8.5 times the amount,and the concentration of artificial sweat ethanol solution was 27%.Under this condition,the comprehensive score was 89.12,the theoretical value was 89.24,and the relative error with the theoretical value was less than 5%.Conclusion:The methods and indexes used in this experiment are suitable for the simultaneous extraction of Chuanxinlian(Andrographis paniculate)and Liaogewang(Wikstroemia indica).The obtained extract has high anti-inflammatory effect,low toxicity and small side effects.
