胃癌P2RX7表达状态与中医脾虚毒蕴证的相关性研究

Correlation Study between Expression Status of P2RX7 in Gastric Cancer and TCM Syndrome of Spleen-Deficiency and Toxin-Accumulation Syndrome

目的:探讨胃癌组织中P2RX7的表达水平与胃癌脾虚毒蕴证的相关性,并探讨胃癌脾虚毒蕴证的危险因素。方法:收集胃癌新发病例的中医四诊资料,填写中医症状调查表,并进行辨证分型,收集患者的手术或胃镜石蜡切片进行免疫组织化学检测,采用χ^(2)检验、Pearson相关性分析和二分类Logistic回归分析方法研究P2RX7表达与胃癌脾虚毒蕴证的关系。结果:本研究共纳入80例胃癌患者。其中男42例,女38例;有肿瘤家族史者21例;P2RX7高表达者47例,低表达者33例;TNM分期为Ⅰ期者4例,Ⅱ期者19例,Ⅲ期者44例,Ⅳ期者13例;中医辨证为脾虚毒蕴证者36例,非脾虚毒蕴证者44例。胃癌脾虚毒蕴证患者的P2RX7蛋白表达水平显著高于胃癌非脾虚毒蕴证患者(P<0.05),Pearson相关性分析显示P2RX7与脾虚毒蕴证存在相关性(R^(2)=0.253,P<0.01),Logistic回归分析显示P2RX7蛋白高表达和肿瘤家族史是胃癌患者脾虚毒蕴证的独立危险因素(P<0.05)。结论:胃癌脾虚毒蕴证患者的肿瘤组织中P2RX7显著高表达,胃癌患者肿瘤组织中P2RX7表达状态与脾虚毒蕴证相关,且P2RX7高表达是胃癌患者脾虚毒蕴证的独立危险因素。

Objective:To analyze the correlation between the expression level of P2RX7 and gastric cancer with spleen-deficiency and toxin-accumulation syndrome,and explore the risk factors of gastric cancer with spleen-deficiency and toxin-accumulation syndrome.Methods:New cases of gastric cancer and the datas of the four diagnosis of traditional Chinese medicine were collected.The TCM symptom questionnaire and syndrome differentiation was completed.The patients'surgical or gastroscope paraffin sections were collected for immunohistochemical detection.Thenχ2 test,Pearson correlation analysis and binary Logistic regression analysis were used to study the relationship between the expression of P2RX7 and the syndrome of spleen-deficiency and toxin-accumulation in gastric cancer.Results:A total of 80 gastric cancer cases were included in this study,with 42 males and 38 females.There were 21 cases with family history of tumors.Totally 47 cases were with high expression of P2RX7 and 33 cases with low expression.There were 4 cases were classified as TNM stage I,19 cases as stageⅡ,44 cases as stageⅢ,and 13 cases as stageⅣ.Totally 36 cases were indentified as spleen-deficiency and toxin-accumulation syndrome and 44 cases were non-spleen-deficiency and toxin-accumulation.The gastric cancer patients with spleen-deficiency and toxic-accumulation syndrome showed higher expression level of P2RX7 protein than those with non-spleen-deficiency and toxic-accumulation syndrome(P<0.05).Pearson correlation analysis showed that P2RX7 had a certain correlation with spleen-deficiency and toxin-accumulation syndrome(R2=0.253,P<0.01),and Logistic regression analysis showed that high expression of P2RX7 protein and family history of tumors were independent risk factors for spleen-deficiency and toxin-accumulation in patients with gastric cancer(P<0.05).Conclusion:The expression of P2RX7 is significantly higher in gastric cancer patients with spleen-deficiency and toxic-accumulation syndrome.The expression status of P2RX7 in tumor tissue of gastric cancer patients is related to spleen-deficiency and toxin-accumulation syndrome,and high expression of P2RX7 is an independent risk factor for spleen-deficiency and toxic-accumulation syndrome in gastric cancer patients.
