不同酶组合致兔眼玻璃体液化的实验研究

摘要 目的探讨透明质酸酶(HA)联合纤溶酶(PL)及软骨素酶(CA)联合基质金属蛋白酶3(MMP-3)诱导兔眼玻璃体液化及玻璃体后脱离(PVD)的快速性、安全性及有效性。方法通过完全随机分组法将24只新西兰白兔分成3组,A组11只,B组11只,C组2只作为空白组,A组和B组白兔均以右眼为实验眼,左眼为对照眼。两组实验眼分别注射20IUHA+1 IUPL及0.2 U CA+10 ng MMP-3于视盘前玻璃体后1/3处,两组对照眼相同部位注射BSS 0.1 ml,C组不做任何处理。注药前后,进行肉眼、裂隙灯显微镜、间接检眼镜和眼部B超检查。注射后3、7、14 d摘除眼球,进行冰冻切片行HE染色观察。结果HE染色显示两组实验眼视网膜结构清晰完整,排列整齐,未见明显视网膜异常。0.2U CA联合10ng MMP-3及20IUHA联合1IUPL应用玻璃体内注射均可以诱导玻璃体液化和完全性PVD的发生,0.2U CA联合10ng MMP-3诱导玻璃体液化及PVD更具快速性且在第7天诱导效果更佳。结论两组药物均对视网膜和其他眼内组织是安全的,均可有效诱导PVD,0.2 U CA联合10ng MMP-3所致兔眼玻璃体液化效果更为快速。 Objective To investigate the rapidity,safety and effectiveness of hyaluronidase(HA)combined with plasmin(PL)and chondroitin enzyme(CA)combined with matrix metalloproteinase 3(MMP-3)in inducing vitreous liquefaction and posterior vitreous detachment(PVD)in rabbits.Methods Twenty-four rabbits were randomly divided into three groups,including eleven rabbits in group A,eleven rabbits in group B and two rabbits in group C as the blank group.One eye of each rabbit in either group A and B were experimental eye and the other eye was control eye.The position of drug injection was in posterior 1/3 of vitreous before papillary.The experimental eyes of group A were treated with 20IU HA and 1IU PL.Also,the experimental eyes of group B were treated with 0.2U CA and 10ng MMP-3.The control group was received equivalent dose of balanced salt solution(BSS).Group C would not accept any processing.Before and after injection,visual examination,slit lamp microscope,indirect ophthalmoscope,Bultrasound examination were performed.Eyes were removed on the 3rd,7th,and 14th day after injection,and frozen sections were performed for HE staining observation.Results HE staining showed that the retinal structures of the two experimental groups were clear and complete,arranged neatly,and no obvious retinal abnormalities were observed.Both 0.2 U CA combined with 10 ng MMP-3 and 20 IU HA combined with 1 IU PL intravitreal injection could induce vitreous liquefaction and complete PVD.0.2 U CA combined with 10 ng MMP-3 induced vitreous liquefaction and PVD more quickly and had better induction effect on the 7th day.Conclusions Both groups of drugs are safe for the retina and other intraocular tissues,and both can effectively induce PVD.The effect of 0.2 U CA combined with 1Ong MMP-3 on vitreous liquefaction in rabbit eyes is taken more quickly.
