摘要
本研究以温泉分离的地衣芽孢杆菌(Bacillus licheniformis)为前体材料,通过一步水热法合成了绿色生物材料(N-CDs),N-CDs具有独特的光学特性、优异的生物相容性和可忽略不计的生物毒性,是生物多色成像的理想荧光染料。利用N-CDs作为生物成像的荧光探针进行实验,结果表明N-CDs能成功染色洋葱细胞的细胞膜结构,不会造成质壁分离;可对酵母细胞的细胞膜进行荧光染色,且酵母细胞可以正常出芽生殖;N-CDs可在植物叶片中通过叶脉和细胞间隙进行扩散成像;也可以正常进入线虫体内进行荧光染色,且不会影响线虫的生命特征。
In this study,N-CDS was successfully synthesized by one-step hydrothermal method with Bacillus Licheniformis isolated from hot spring as a precursor material.The obtained N-CDs has unique optical properties,excellent biocompatibility and negligible biotoxicity and is an ideal fluorescent dye for biological polychromatic imaging.N-CDs was used as a fluorescent probe for biological imaging.The result of cell imaging showed that N-CDs could successfully stain the cell membrane structure of onion inner cells without causing plasma-wall separation.The imaging of microbial experiments showed that N-CDs could fluorescein stain Saccharomyces cerevisiae and S.cerevisiae still could sprout and reproduce normally.The results of plant leaf imaging showed that N-CDs could be diffused in plant leaves through veins and intercellular space.The biological imaging results of Caenorhabditis Elegans showed that N-CDs could enter the body normally for fluorescence staining without affecting the vital signs.
