糖尿病肾病大鼠血清NSE、S100B相关性实验研究

摘要 目的:探究神经元特异性烯醇化酶(NSE)、S100B蛋白(S100B)在糖尿病肾病大鼠中的表达与诊断价值。方法:将40只SD雄性大鼠随机分为空白组10只、模型组(DN组)30只,将DN组适应性喂养7天,造模前禁食12 h,按规定剂量于大鼠左下腹腹腔注射链脲佐菌素(60 mg/kg),72 h后尾部采血测得大鼠随机血糖≥16.7 mmol/L,即为糖尿病大鼠。检测大鼠24h尿微量白蛋白(mALB)﹥30 mg/L后得到糖尿病肾病大鼠模型。于第10周处死大鼠,用酶联免疫法(ELISA)检测大鼠血清NSE、S100B的表达量。结果:DN组大鼠血清中NSE、S100B表达明显高于空白组,差异有统计学意义(P﹤0.05);DN组大鼠血清NSE与mALB表达相关性分析无统计学意义(P﹥0.05);DN组大鼠血清S100B与mALB呈正相关(P﹤0.01);DN组大鼠血清NSE与血糖值呈正相关(P﹤0.05),DN组大鼠血清S100B与血糖值呈正相关(P﹤0.01);DN组大鼠血糖值与mALB表达呈正相关(P﹤0.01)。结论:血清NSE、S100B对糖尿病肾病有一定的辅助诊断价值。 Objective:To approach the expressions and diagnostic values of neuron specific enolase(NSE)and S100B protein S100B in rats with diabetes nephropathy.Methods:40 male SD rats were randomly divided into blank group(10 rats)and model group(DN group,30 rats).DN group was adaptively fed for 7 days,fasted for 12 hours before modeling,and injected streptozotocin(60mg/kg)into the left lower abdominal cavity of rats by the prescribed dose.After 72 hours,the blood samples were taken from the tail;the measured random blood sugar of rats was≥16.7mmol/L,which showed that the rats were diabetes rats.The rat model of diabetes nephropathy was obtained after the detected 24h urinary microalbumin(mALB)of rats was>30mg/L.At the 10th week,the rats were euthanized,and the expression levels of serum NSE and S100B in rats were measured by enzyme-linked immunosorbent assay(ELISA).Results:The expressions of NSE and S100B in the serum of rats in DN group were significantly higher than those in blank group,and the difference was statistically significant(P﹤0.05).The analysis of correlativity between NSE and mALB expressions in serum of rats of DN group was no statistical significance(P﹥0.05).S100B in serum of rats of DN group was positively correlated with mALB(P﹤0.01).NSE in serum of rats of DN group was positively correlated with blood glucose level(P﹤0.05).S100B in serum of rats of DN group was positively correlated with blood glucose levels(P﹤0.01).The blood glucose level of rats in DN group was positively correlated with mALB expression(P﹤0.01).Conclusion:Serum NSE and S100B have certain auxiliary diagnostic value for diabetes nephropathy.
