摘要
[目的]找到一种快速、高通量、低成本提取油菜DNA的方法。[方法]将改良碱煮法快速高通量提取油菜DNA的方法与试剂盒法(磁珠法)提取的DNA质量进行比较,并进一步分析油菜植株不同部位和叶片不同时期的提取效果。[结果]与以磁珠法为代表的传统提取法相比较,改良的碱煮法快速高通量提取油菜DNA的方法,用于普通的SSR分子标记检测没有明显差异。同时,通过对油菜植株不同部位和叶片不同时期的提取效果进行分析,发现没有明显差异,表明该方法在分子标记辅助育种以及DNA纯度鉴定等试验中可广泛应用。[结论]改良的碱煮法提取DNA可以在油菜不同时期和不同部位应用,进一步提高了碱煮法的使用效率。
[Objective]The aim of this study was to find a rapid,high-throughput and low-cost method for extracting rapeseed DNA.[Method]The quality of DNA extracted by improved alkali boiling method and the reagent kit method(magnetic bead method)was compared,and the extraction effects of different parts of plants and leaves at different stages were further analyzed.[Result]Compared with the traditional extraction method represented by magnetic bead method,the improved alkali boiling method for rapid and high-throughput extraction of rapeseed DNA had no significant difference in the detection of ordinary SSR molecular markers.At the same time,by analyzing the extraction effects of different parts of plants and different stages of leaves,it was found that there was no significant difference,indicating that this method can be widely used in molecular marker-assisted breeding and DNA purity identification experiments.[Conclusion]The improved alkali boiling method for DNA extraction can be applied in different periods and different parts of rapeseed,which further improves the efficiency of alkali boiling method.
