摘要
【目的】探究绵羊(Ovis aries)RFXANK基因多态性及其与肉用性状之间的关系,为优质肉羊育种提供有效的分子遗传标记。【方法】采用液相捕获技术测定杜泊羊(D)、小尾寒羊(XH)和滩羊(T)3组91头羊,通过液相捕获技术获取RFXANK的突变位点,并从中选出3个多态位点(rs160009883、rs420498050、rs405773959)。针对筛选出的位点,采用飞行质谱检测技术对杜滩寒三元杂交羊(DTH)、杜泊羊、小尾寒羊和滩羊共30只进行遗传分型,并与其肉用性状进行关联分析。【结果】rs160009883位点AA基因型宰前活重、胴体重、净肉重极显著高于CC基因型(P<0.01),AA基因型净肉率显著高于CC基因型(P<0.05),CA基因型净肉重显著高于CC基因型,AA、CA基因型胸围显著高于CC基因型;rs420498050和rs405773959位点AA基因型宰前活重、屠宰率、净肉重、胴体重、净肉率极显著高于GG基因型,AA基因型胸围、背膘厚显著高于GG基因型;3个位点不同基因型与GR值、肌纤维直径、肌纤维密度、蛋白质、肌苷酸、丙氨酸和缬氨酸均呈显著相关,与各内脏器官重量差异显著。【结论】RFXANK基因的3个位点对绵羊肉用性状具有显著影响,可作为绵羊肉用性状的分子标记之一,为提高杜泊羊、小尾寒羊和滩羊群体肉用性状提供参考。
【Objective】The present paper aimed to investigate the polymorphism of RFXANK gene in sheep(Ovis aries)and its relationship with meat performance to provide effective molecular genetic markers for the breeding of high quality meat sheep.【Method】A total of 91 sheep from three populations,Dupo(D),Little-tailed sheep(XH)and Tandan sheep(T),were used to obtain the variant loci of RFXANK gene using liquid capture sequencing technology.A total of 30 sheep from Dupo(D),Tandoori(T),Little Tail(XH)and Dutang Cold Ternary(DTH)crosses were genotyped and correlated with their meat performance using flight mass spectrometry.【Result】The rs160009883 locus AA genotype was highly significantly higher than the CC genotype in live weight before slaughter,carcass weight and net meat weight(P0.01),the AA genotype was significantly higher than the CC genotype in net meat percentage(P0.05),the CA genotype was significantly higher than the CC genotype in net meat weight,and the AA and CA genotypes were significantly higher than the CC genotype in chest circumference.Live weight before slaughter,slaughter rate,net meat weight,carcass weight and net meat rate were highly significant for AA genotypes than GG genotypes at rs420498050 and rs405773959 loci,and chest circumference and backfat thickness were significantly higher for AA genotypes than GG genotypes;Different genotypes at three loci were associated with GR value,muscle fibre diameter,muscle fibre density,protein,inosine,alanine and valine were significantly correlated with the weight of each visceral organ.【Conclusion】The three loci of RFXANK gene have significant effects on meat performance in sheep and can be used as one of the molecular markers for meat performance in sheep,providing a reference for improving meat performance in the Dupo,Little-tailed sheep and Tandan sheep population.
