摘要
试验为研究玉米须总黄酮的提取工艺及其抗氧化活性,采用离子液体-微波协同提取玉米须中的总黄酮。在单因素试验的基础上,应用响应面法优化提取工艺。结果显示,最佳工艺为微波功率450 W、离子液体浓度0.65 mol/L、液料比25 mL/g、提取时间30 min。在此条件下,总黄酮提取量为5.21 mg/g。体外抗氧化试验结果表明,玉米须中提取的黄酮类物质的抗氧化能力较好,对羟基自由基和1,1-二苯基-2-三硝基苯肼(DPPH)自由基的半数抑制浓度(IC50)分别为146.7142、120.4200 mg/L。研究表明,响应面法优化离子液体-微波协同提取玉米须总黄酮稳定、可行,可为植物活性成分在动物生产领域的应用提供参考。
The experiment was to study the extraction process of total flavonoids from corn silk and their antioxidant activity,ionic liquid-microwave assisted extraction was used to extract total flavonoids from corn silk.Based on singlefactor methodology,the response surface method was used to optimize the experimental conditions.The results showed that the optimum conditions were ultrasonicwave power 450 W,ionic liquid concentration 0.65 mol/L,liquid to solid ratio 25 mL/g,microwave time 30 min.Under the above conditions,the isoflavones yield of the corn silk was 5.21 mg/g.The results of antioxidant activity experiments showed that the flavonoids extracted from corn whisker had good antioxidant capacity.The corresponding IC50 values of hydroxyl radical and DPPH radical were 146.7142 and 120.4200 mg/L.The study indicates that extracting total flavonoids from corn silk by ionic liquid-microwave assisted extraction method is stably and feasibly,which can provide reference for the application of active ingredients from plants in the field of animal production.
