棉酚对食管癌细胞系SEG-1细胞裸鼠模型体内作用的研究

摘要 目的探讨棉酚在食管癌细胞系SEG⁃1细胞裸鼠模型体内的作用。方法取健康SPF级BALb/c雌性裸鼠30只,分为6组,即醋酸棉酚40 mg/kg组、醋酸棉酚60 mg/kg组、单醛棉酚40 mg/kg组、单醛棉酚60 mg/kg组、空白对照组、阴性对照组,每组5只,空白组未接种及棉酚处理,其余5组均接种SEG⁃1细胞,除阴性对照组外的4组分别在接种肿瘤3 d后开始给予相应剂量棉酚处理,观察各组肿瘤体积和质量的变化,计算肿瘤生长抑制率和肿瘤生长延迟时间,观察裸鼠接种后体质量变化,解剖测量肝、脾质量的变化。结果醋酸棉酚40 mg/kg组、醋酸棉酚60 mg/kg组、单醛棉酚40 mg/kg组、单醛棉酚60 mg/kg组肿瘤生长抑制率分别为14.3%、31.2%、33.7%、32.9%(P<0.05)。给予40 mg/kg醋酸棉酚干预的接种裸鼠肝脏质量明显低于阴性对照组,而给予40 mg/kg或60 mg/kg单醛棉酚干预的裸鼠肝脏较对照组并没有减轻。醋酸棉酚40 mg/kg组、醋酸棉酚60 mg/kg组裸鼠生存时间明显短于阴性对照组。结论醋酸棉酚、单醛棉酚均能抑制食管癌细胞系SEG-1裸鼠模型体内肿瘤的生长。单醛棉酚对裸鼠模型的毒性作用比醋酸棉酚小。 Objective:To investigate the effect of gossypol on SEG-1cells in nude mice model of esophageal carcinoma.Methods:Thirty healthy SPF female BALb/c nude mice were divided into 6 groups,namely gossypol acetic acid 40 mg/kg group,gossypol acetic acid 60 mg/kg group,mono-aldehyde gossypol 40 mg/kg group,mono-aldehyde gossypol 60 mg/kg group,blank control group and negative control group.There were 5 rats in each group.The blank group did not receive inoculation and gossypol treatment.The remaining 5 groups were inoculated with SEG-1 cells and then given corresponding concentration gossypol after 3 days after tumor inoculation.Then the variation of tumor volume and weight were observed,the tumor growth delay and tumor growth inhibition rate were calculated,the body weight changes after inoculation were observed,and at last the weight of liver and spleen was measured.Results:All subcutaneous mass with drug treat reduced in different levels and the growth rate of the mass slowed down.Inhibition rates of tumor growth of gossypol acetic acid 40 mg/kg,gossypol acetic acid 60 mg/kg group,mono-aldehyde gossypol 40 mg/kg group,and mono-aldehyde gossypol 60 mg/kg group were 14.3%,31.2%,33.7%,32.9%respectively(P<0.05).The liver weight in acetic acid gossypol 40 mg/kg was significantly lower than that of the negative control group,while the nude mice liver of mono-aldehyde gossypol 40 or 60 mg/kg group did not reduce compared with that of the negative control group.The survival time of gossypol acetic acid 40 mg/kg and 60 mg/kg group was significantly shorter than that of the negative control group.Conclusion:Gossypolacetic acid and mono-aldehyde gossypol can inhibit the tumor growth of esophageal carcinoma in nude mice.The toxic effects of monoaldehyde gossypol on esophageal cancer in nude mice model are less than those of gossypol acetic acid.
