摘要
环状RNA(circular RNA,circRNA)是与疾病发生、发展密切相关的一类非编码RNA,其为共价闭合的环状结构,与线性RNA相比稳定性更好,有望成为疾病检测、诊断的新型指标。circRNA的检测,尤其是定量检测对疾病的诊断十分重要。目前,以核酸扩增为基础的检测是circRNA定量最主要的方法,因此本文总结了以核酸扩增为基础的circRNA检测方法,主要包括以PCR和等温扩增技术为基础的扩增方法,期望为circRNA检测提供理论依据。
Circular RNA(circRNA)discovered in recent years is a type of non-coding RNA and closely related to disease occurrence and development.Compared with linear RNA,circRNA has a more stable covalent closed circular structure,making it a novel biomarker for the detection and diagnosis of diseases.The detection of circRNA,especially quantitative detection,is essential for the diagnosis of diseases.Currently,the main method for the quantification of circRNA is based on nucleic acid amplification.In this paper,we summarize the amplification methods of circRNA detection based on nucleic acid amplification,mainly including polymerase chain reaction(PCR)and isothermal amplification,expecting to provide a theoretical basis for circRNA detection.
