玉龙雪山乌鸡mtDNA D-loop区遗传多样性分析 被引量:1

摘要 为阐明玉龙雪山乌鸡的遗传背景信息,试验采用线粒体DNA D-loop区序列为遗传标记,对采自核心产区的55只玉龙雪山乌鸡样品进行群体变异检测,并将所得数据与已发表的云南12个地方鸡种数据进行联合分析。结果显示:在所有个体中共检测到25个多态位点,包含单一信息位点2个,简约信息位点23个,核苷酸替换主要为转换,转换与颠换比为6.40。在该群体中共定义了14种单倍型,其单倍型数多样度为0.867±0.027,核苷酸多样度为0.01074±0.00114,群体内平均遗传距离为0.011±0.003。玉龙雪山乌鸡与云南其他地方鸡群体间的平均遗传距离在0.013(宁蒗高原鸡)~0.019(大围山微型鸡)之间,遗传分化系数Fst值在0.078(宁蒗高原鸡)~0.543(兰坪绒毛鸡)之间。在世系组成上,玉龙雪山乌鸡中含有B、E、F和G世系,占比分别为20.00%、3.64%、9.09%和67.27%,总体上玉龙雪山乌鸡群体中云南及周边地区的地方鸡血源在76.36%以上。结果揭示,玉龙雪山乌鸡的遗传多样性相对丰富,与云南其他地方鸡种间存在不同程度的遗传分化,其母系世系组成与云南其他地方鸡有较大差异。 In order to clarify the genetic background information of Yulong Snow Mountain black-bone chicken,55 samples of Yulong Snow Mountain black-bone chicken from its core production area were detected for population variation by using mitochondrial DNA D-loop sequence as genetic marker,and the data were analyzed jointly with the published data of 12 other local chicken breeds in Yunnan.A total of 25 polymorphic sites were detected in all individuals,including 2 single information sites and 23 parsimony information sites,and nucleotide substitutions were mainly conversion,with a conversion to inversion ratio of 6.40.A total of 14 haplotypes were defined in this chicken,and the haplotype diversity,nucleotide diversity and mean genetic distance within population were 0.867±0.027,0.01074±0.00114 and 0.011±0.003,respectively.The mean genetic distance between Yulong Snow Mountain black-bone chicken and other Yunnan indigenous chickens ranged from 0.013(Ninglang plateau chicken)to 0.019(Daweishan mini chicken),and the Fst value ranged from 0.078(Ninglang plateau chicken)to 0.543(Lanping silky chicken).In terms of lineage composition,this chicken contains B,E,F and G lineages,accounting for 20.00%,3.64%,9.09%and 67.27%,respectively,and in general,its indigenous chicken lineages from Yunnan and its surrounding areas were more than 76.36%.The results indicated that the genetic diversity of Yulong Snow Mountain black-bone chicken was relatively rich,and there were different degrees of genetic differentiation between Yulong Snow Mountain black-bone chicken and other local chicken breeds in Yunnan.And the maternal lineage composition of Yulong Snow Mountain black-bone chicken was also significantly different from that of other local chickens in Yunnan.
