建立超高效液相色谱法测定莫匹罗星软膏含量

摘要 目的:建立莫匹罗星软膏含量测定超高效液相色谱法,提高检测效率。方法:采用超高效液相色谱法(Ultra performance liquid chromatography,UPLC),色谱柱为ACQUITY UPLC BEH C_(18)(2.1 mm×100 mm,1.7μm);流动相为0.01 mol·L^(-1)磷酸二氢钠(1 mol·L^(-1)NaOH调节pH至6.3)-乙腈(75∶25);检测波长为230 nm;流速为0.4 mL·min-1。结果:建立了莫匹罗星软膏含量测定超高效液相色谱方法,此方法精密度好,准确度高,其线性范围为0.01~1 mg·mL^(-1),R 2为0.9996。结论:相比于高效液相色谱(High Performance Liquid Chromatography,HPLC)法,本文所提方法更便捷高效,可有效地控制莫匹罗星软膏含量。 Objective:The aim of this study is to establish an ultra performance liquid chromatography method for the determination of mupirocin ointment and to improve the detection efficiency.Methods:In this study,ultra performance liquid chromatography(UPLC)method was used with the ACQUITY UPLC BEH C_(18)(2.1 mm×100 mm,1.7μm)as the chromatographic column.In this process,the mobile phase,detection wavelength and flow rate were 0.01 mol·L^(-1)sodium dihydrogen phosphate(adjusted pH to 6.3 by 1 mol·L^(-1)NaOH)-acetonitrile(75∶25),230 nm and 0.4 mL·min-1,respectively.Results:An ultra performance liquid chromatography method with good precision and high accuracy for the determination of mupirocin ointment content was established with the linear range was 0.01-1 mg·mL^(-1)and the R 2 was 0.9996.Conclusion:Compared with High Performance Liquid Chromatography(HPLC),the proposed method is more convenient and efficient,and can effectively control the content of mupirocin ointment.
