miR-133a抑制增生性瘢痕成纤维细胞增殖和转移的实验研究

摘要 目的探讨mi R-133a对增生性瘢痕成纤维细胞增殖和转移的作用。方法自2016年6月至2020年6月,选取北部战区总医院烧伤整形科收治的30例确诊为增生性瘢痕患者的增生性瘢痕组织样本及30例正常皮肤瘢痕组织的标本,通过实时荧光定量PCR检测miR-133a的表达水平。然后,将购买的增生性瘢痕成纤维细胞分为3组,采用miR-133a、miR-133a模拟物、miR-133a抑制物转染后,实时荧光定量PCR检测miR-133a表达情况,使用细胞计数试剂盒-8(cell counting kit-8assay,CCK-8)检测miR-133a对增生性瘢痕成纤维细胞增殖的调节作用。通过Transwell检测miR-133a对增生性瘢痕成纤维细胞迁移和侵袭的调节作用。结果miR-133a在增生性瘢痕组织中的表达显著低于正常瘢痕组织。成功构建过表达/低表达mi R-133a的增生性瘢痕成纤维细胞后,CCK-8和Transwell结果显示,miR-133a上调能够抑制细胞的增殖、迁移和侵袭,miR-133a下调促进增生性瘢痕成纤维细胞的增殖、迁移和侵袭。结论miR-133a在增生性瘢痕组织中表达下调,能够抑制增生性瘢痕成纤维细胞的增殖和转移。 Objective To investigate the effect of miR-133a on the proliferation and metastasis of hypertrophic scar fibroblasts.Methods From June 2016 to June 2020,Department of Burn and Plastic Surgery of General Hospital of Northern Theater Command selected 30 hypertrophic scar tissue samples and 30 normal skin scar tissue samples.The expression of miR-133a was detected by real-time PCR.After transfection of miR-133a,miR-133a mimic,miR-133a inhibitor into hypertrophic scar fibroblasts,real-time PCR was used to detect the expression of miR-133a.Cell counting kit-8 assay(CCK-8)was used to detect the regulatory effect of miR-133a on the proliferation of hypertrophic scar fibroblasts.The effect of miR-133a on the migration and invasion of hypertrophic scar fibroblasts was detected by transwell.Results The expression of miR-133a in hypertrophic scar tissue was significantly lower than that in normal scar tissue.After successfully constructing hypertrophic scar fibroblasts with overexpression or low expression of miR-133a,CCK-8 and transwell results showed that upregulation of miR-133a could inhibit the proliferation,migration and invasion of hypertrophic scar fibroblasts,while downregulation of miR-133a could promote the proliferation,migration and invasion of hypertrophic scar fibroblasts.Conclusion Downregulation of miR-133a expression in hypertrophic scar tissue can inhibit the proliferation and metastasis of hypertrophic scar fibroblasts.
