基于OSMAC策略对淮河来源真菌Talaromyces stipitatus BMC-16化学成分多样性的研究

摘要 目的采用单菌株多次级代谢产物(OSMAC)策略对分离自安徽蚌埠淮河区域泥样的真菌Talaromyces stipitatus BMC-16进行化学多样性的研究。方法基于OSMAC策略,选用19种不同培养基分别小规模发酵培养菌株Talaromyces stipitatus BMC-16。观察不同培养基条件下菌株生长状态,采用高效液相色谱法(HPLC)和薄层色谱法(TLC)分析小规模发酵次级代谢产物化学多样性,纸片法检测其抑菌活性。结果在不同培养基中,菌株的产量、生长形态、代谢产物分布以及抑菌活性均有差异,显示出较佳的代谢潜能。对不同培养基发酵物中的主峰进行分离纯化,获得化合物1,经鉴定为已知化合物stipitatic acid。结论基于OSAMC策略,改变培养基条件,对菌株Talaromyces stipitatus BMC-16的次级代谢产物有显著的影响,丰富了该菌株次级代谢产物的化学多样性。 Objective To study the chemical diversity of Talaromyces stipitatus BMC-16 isolated from soil samples collected from the Huai river in Bengbu,Anhui province using one strain many compounds(OSMAC)strategy.Methods Nineteen different media were selected for small-scale fermentation of the strain Talaromyces stipitatus BMC-16 based on the OSMAC strategy.The growth status of the strain under different media conditions were observed.The chemical diversity was analyzed by high performance liquid chromatography(HPLC)and thin layer chromatography(TLC).The antibacterial activities of the secondary metabolites were detected by paper sheet method.Results The yield,the growth morphology,the chemical diversity,and the antibacterial activity of the secondary metabolites were distinguished in different media,and the strain showed better metabolic potential.Compound 1(the main peak in the fermentation of different media)was isolated and identified as the known compound,stipitatic acid.Conclusion Based on OSMAC strategy,the medium conditions have a significant effect on the chemical diversity of secondary metabolites of the fungus Talaromyces stipitatus BMC-16,and the chemical diversity of secondary metabolites of the strain is enriched.
