加味参芪地黄汤对糖尿病肾病大鼠足细胞自噬的影响

Effect of Modified Shenqi Dihuang Decoction(加味参芪地黄汤)on Podocyte Autophagy in Diabetic Nephropathy Rats

目的:观察加味参芪地黄汤对糖尿病肾病(diabetic nephropathy,DN)大鼠足细胞自噬的影响。方法:从50只SD大鼠中取10只大鼠作正常对照组,其余采用高糖高脂饲料+STZ诱导DN大鼠模型,造模成功后将DN大鼠分为4组,分别为模型组、阳性对照组(洛丁新,0.90 mg/kg)、加味参芪地黄汤高剂量组(44.10 g/kg)、加味参芪地黄汤低剂量组(11.03 g/kg),每组9只,各组按相应剂量药物灌胃给药,模型组、正常组均予以等体积生理盐水灌胃,每天1次,共8周。HE、Masson染色观察肾脏病理改变;采用免疫组织化学法检测肾组织中自噬蛋白5(Autophagy-related protein 5,Atg5)、微管相关蛋白1轻链3(Microtubule-associated protein 1-light chain 3,LC3)表达情况。结果:肾脏HE染色和MASSON染色结果显示,加味参芪地黄汤可以改善DN大鼠肾小球系膜基质弥漫增生,基底膜增厚、肾小管空泡变性脂肪变性和间质淋巴增生的病理损伤,与阳性对照组相比较有明显差异(P<0.05)。免疫组化结果显示,加味参芪地黄汤可以增强DN大鼠肾脏足细胞自噬蛋白Atg5、LC3的表达,与阳性对照组相比有明显差异(P<0.05)。结论:加味参芪地黄汤能够改善DN大鼠肾组织病理损伤,促进DN大鼠的足细胞自噬。

Objective:To observe the effect of Modified Shenqi Dihuang Decoction on podocyte autophagy in rats with diabetic nephropathy(DN).Methods:40 male SD rats were fed with high sugar and high fat diet+STZ to induce DN rat models.After successful modeling,the DN rats were divided into 4 groups:model group,positive control group(Benazepril Hydrochloride,0.90 mg/kg),Modified Shenqi Dihuang Decoction high dose group(44.10 g/kg),Modified Shenqi Dihuang Decoction low dose group(11.03 g/kg),9 rats in each group,10 rats were taken as normal control group,each group rats were given the corresponding medicine once a day for 8 weeks.The pathological changes of kidney tissue were observed by HE and Masson staining.The expressions of Atg5 and LC3 in renal tissues were detected by immunohistochemistry.Results:HE staining and Masson staining showed that the Modified Shenqi Dihuang Decoction could improve the pathological changes of glomerular mesangial matrix diffuse proliferation,basement membrane thickening,tubular vacuolar degeneration and steatosis and interstitial lymphatic hyperplasia in DN rats,which were significantly different from those in the positive control group(P<0.05).The Modified Shenqi Dihuang Decoction could increase the expressions of Atg5 and LC3 in renal podocytes of DN rats,and there were significant differences compared with the positive control group(P<0.05).Conclusion:Modified Shenqi Dihuang Decoction can improve the pathological damage of DN rats,promote podocyte autophagy.
