基于脑肠互动探讨眼针调节Th17/Treg免疫平衡治疗溃疡性结肠炎作用机制研究 被引量:1

摘要 目的从脑肠互动角度探讨眼针治疗溃疡性结肠炎(ulcerative colitis,UC)的疗效作用机制。方法50只SD大鼠随机分为空白对照组、模型对照组、西药治疗组、眼针治疗组、针药联合组,每组10只。采用5%DSS水溶液自由饮用7 d建立UC大鼠模型,每日灌胃、眼针(肺-大肠区)治疗1次,连续10 d。观察一般情况及每日DAI评分,处死后行HE染色观察结肠组织病理学变化,ELISA法检测血清、脑组织中白细胞介素(IL)-6、TGF-β、IL-17、IL-10含量,q-PCR检测结肠黏膜、脑组织中FoxP3、RORγt mRNA含量,Western blotting法检测结肠黏膜、脑组织中FoxP3、RORγt蛋白表达。采用SPSS 19.0进行统计学分析。结果与模型对照组相比,西药对照组、眼针治疗组和针药联合组DAI评分降低,病理损伤明显减轻,结肠组织与脑组织中IL-6、IL-17、TGF-β、RORγt表达水平均降低,IL-10、FoxP3表达均增加,差异有统计学意义(P<0.05)。针药联合组与眼针治疗组在(IL)-6、TGF-β、IL-17、IL-10、FoxP3、RORγt表达方面有统计学意义(P<0.05)。结论眼针能通过脑肠互动调节细胞因子的水平、调控FoxP3与RORγt表达以恢复Th17/Treg免疫平衡,从而可以改善UC大鼠的临床症状,修复结肠黏膜损伤。 Objective To investigate the therapeutic effect of eye acupuncture on ulcerative colitis from the perspective of brain-gut interaction.Methods 50 SD rats were randomly divided into blank control group,model control group,Western medicine treatment group,eye acupuncture treatment group and acupuncture combined group,with 10 rats in each group.The UC rat model was established by drinking 5%DSS solution freely for 7 days.The rats were treated by gavage and eye needle(lung-large intestine area)once a day for 10 consecutive days.After death,HE staining was performed to observe the pathological changes of colon.ELISA was used to detect the contents of interleukin IL-6,TGF-β,IL-17 and IL-10 in serum and brain tissue,q-PCR was used to detect the mRNA contents of FoxP3 and RORγt in colon mucosa and brain tissue.The expressions of FoxP3 and RORγt mRNA in colon mucosa and brain were detected by Western blotting.SPSS19.0 was used for statistical analysis.Results Compared with the model control group,the DAI score of the Western medicine control group,the eye acupuncture treatment group and the acupuncture combined group were significantly reduced,the expression levels of IL-6,IL-17,TGF-β and RORγt were decreased,and the expression levels of IL-10 and FoxP3 were increased in colon tissue and brain tissue,and the differences were statistically significant(P<0.05).There were significant differences in the expressions of IL-6,TGF-β,IL-17,IL-10,FoxP3,RORγt between the acupuncture combined group and the eye acupuncture group(P<0.05).Conclusion Eye acupuncture can restore Th17/Treg immune balance by regulating the level of cytokines and the expression of FoxP3 and RORγt through brain-gut interaction,which can improve the clinical symptoms of UC rats and repair colonic mucosal injury.
