金匮温经汤对妇科虚寒证模型大鼠子宫内膜血管损伤的影响

Effect of Jingui Wenjing Decoction(金匮温经汤)on Endometrial Vascular Injury in Rats with Gynecology Deficiency Cold Syndrome

目的观察金匮温经汤对妇科虚寒证模型大鼠子宫VEGF、LIF、STAT、P-STAT3表达的影响,从子宫内膜的修复作用探讨金匮温经汤治疗妇科虚寒证的作用靶点。方法60只3月龄SD雌性清洁级大鼠,随机数字表法分为3组,对照组、模型组、治疗组(各20只)。模型组、治疗组采用延长冰箱冷冻加冰水浸泡时间的方法造模,治疗组造模同时给予金匮温经汤灌胃,对照组、模型组给予同体积蒸馏水灌胃,连续28 d。处死大鼠,取子宫,采用免疫组织化学染色和western blot检测VEGF、LIF、STAT、P-STAT3的表达。结果与对照组比较,模型组大鼠子宫子宫腔狭窄或增大,内膜层腺体、血管稀疏,子宫内膜变薄(P<0.05),VEGF表达升高(P<0.05),LIF、STAT3、P-STAT3的蛋白表达降低(P<0.05);与模型组比较,治疗组内膜腺体较多,分布均匀,内膜层血管丰富,内膜增厚(P<0.05),VEGF的表达降低,LIF、STAT3、P-STAT3的蛋白表达升高(P<0.05)。结论金匮温经汤通过下调子宫内膜VEGF的表达,上调LIF、STAT3、P-STAT3的蛋白表达水平,增加子宫内膜厚度,修复虚寒证模型大鼠子宫内膜的损伤,治疗妇科虚寒证。

Objective To observe the effect of Jingui Wenjing Decoction(金匮温经汤)on the expressions of protein vascular endothelial growth factor(VEGF),LIF,signal transducer and activator of transcription(STAT)and phosphorylation signal transducer and activator of transcription 3(P-STAT3)in rats endometrial with deficiency cold syndrome.To explore the therapeutic target of Jingui Wenjing Decoction treating gynecological deficiency cold syndrome from the repairing effect of endometrium.Methods Sixty SD female clean rats of 3 months old were randomly divided into 3 groups:control group,model group,treatment group(20 in each).The model group and the treatment group were made by extending the freezer and ice water soaking time.At the same time,the treatment group were given Wenjing Decoction,and the control group and model group were given the same volume of distilled water for 28 days.The rats were sacrificed,and the uterus were taken out.The expressions of VEGF,LIF,STAT and P-STAT3 were detected by immunohistochemical staining and Western blot.Results Compared with the control group,the model group rats had narrow or enlarged uterine lumen,sparse endometrial glands and blood vessels,thinning of the endometrium(P<0.05),increased VEGF expression(P<0.05),and decreased protein expressions of LIF,STAT3 and P-STAT3(P<0.05).Compared with the model group,the treatment group had more endometrial glands with uniform distribution,abundant blood vessels in the endometrial layer,thickening of the endometrium(P<0.05),decreased expression of VEGF and increased protein expression of LIF,STAT3 and P-STAT3(P<0.05).Conclusion Jingui Wenjing Decoction can up-regulate the expressions of LIF,STAT3 and P-STAT3 and reduce VEGF,increase endometrium thickness and repair the endometrium damage of deficiency cold syndrome model rats so as to treat gynecological deficiency cold syndrome.
