枸杞多糖对衰老小鼠MCE细胞高糖损伤后氧化应激的影响

Effect of LBP on oxidative stress after high glucose injury in MCE cells

摘要 目的:研究高糖环境下枸杞多糖(LBP)干预衰老小鼠角膜上皮(MCE)细胞后对氧化应激指标(ROS、SOD1)的影响。方法:根据培养基中葡萄糖浓度将MCE细胞分为五组培养48 h:①葡萄糖5.5 mmol/L组;②葡萄糖25 mmol/L组;③葡萄糖200 mmol/L组;④葡萄糖200 mmol/L+LBP 200μg/ml组;⑤葡萄糖(200 mmol/L)+甘露醇(75 mmol/L)组,用免疫荧光染色各组细胞,使用image J定量分析ROS的具体数值,并用酶联免疫吸附试验(ELISA)检测各组培养基上清中SOD1的含量,进行统计学分析。结果:①与各组比较,葡萄糖浓度200 mmol/L组ROS水平最高(P<0.01),葡萄糖200 mmol/L+LBP 200μg/ml组ROS水平低于葡萄糖浓度200 mmol/L组(P<0.01)。②与各组比较,葡萄糖浓度200 mmol/L组SOD1水平最低(P<0.01),葡萄糖200 mmol/L+LBP 200μg/ml组SOD1水平高于葡萄糖浓度200 mmol/L组(P<0.01)。结论:LBP通过降低氧化应激中ROS水平,升高SOD1水平,保护高糖损伤的MCE细胞。 Objective To study the effects of LBP on oxidative stress indicators ROS,SOD1 after intervention withmouse corneal epithelial(MCE)cells in a high glucose environment.Method MCE cells were divided into five groups according to the concentration of glucose in the culture medium for 48 h:①glucose 5.5 mmol/L(group);②glucose 25 mmol/L(group);③glucose 200 mmol/L(group);④glucose 200 mmol/L+LBP 200μg/ml(group);⑤glucose(200 mmol/L)+mannitol(75 mmom/L)(group),cells in each group were stained with immunofluorescence,and specific values of ROS were quantified using image J.The content of SOD1 in the supernatant of the culture medium via each group was detected by enzyme-linked immunosorbent assay(ELISA)and statistically analyzed.Results①Compared with each group,the highest ROS level was found in the glucose concentration 200 mmol/L group(P<0.01),and the ROS level was lower in the glucose concentration 200 mmol/L+LBP 200μg/ml(group)than in the glucose concentration 200 mmol/L group(P<0.01).②Compared with each group,the lowest SOD1 level was found in the glucose concentration 200 mmol/L group(P<0.01),and SOD1 levels were higher in the glucose 200mmol/L+LBP 200μg/ml(group)than in the glucose concentration 200 mmol/L group(P<0.01).Conclusion LBP protects MCE cells damaged by high glucose by decreasing ROS levels and elevating SOD1 levels in oxidative stress.
