摘要
目的 探讨黄连素对高盐高糖诱发的秀丽隐杆线虫形态和寿命改变的干预作用。方法 取秀丽线虫进行培养,随机分为正常对照组、高盐组、高糖组、高盐高糖组。正常对照组加入线虫NGM培养基;高盐组、高糖组、高盐高糖组下设模型对照组、10 mg/L黄连素干预组、20 mg/L黄连素干预组,先分别加入高盐培养基(含0.7%氯化钠的线虫NGM培养基)、高糖培养基(含1%葡萄糖的线虫NGM培养基)、高盐高糖培养基(含0.7%氯化钠和1%葡萄糖的线虫NGM培养基),然后向黄连素干预组中加入相应浓度的黄连素溶液,培养6 d。于第1、3、6天测定各组线虫体长、体宽及寿命,培养第6天采用RT-qPCR法检测线虫类似人胰岛素代谢相关基因daf-2、daf-16 mRNA。结果与正常对照组相比,高盐、高糖以及高盐高糖组线虫体宽增加、寿命缩短(P均<0.05),体长无明显变化(P均>0.05),daf-2 mRNA表达升高、daf-16 mRNA表达降低(P均<0.05);与高盐、高糖以及高盐高糖组相比,添加黄连素组线虫的体宽降低、寿命延长(P均<0.05),体长无明显变化(P均>0.05),daf-2 mRNA表达降低、daf-16 mRNA表达升高(P均<0.05)。结论 黄连素可抑制高盐高糖诱发的秀丽线虫形态改变以及寿命缩短,其机制可能是通过下调daf-2 mRNA表达、上调daf-16 mRNA表达来发挥作用。
Objective To explore the intervention effect of berberine on changes of morphology and lifespan of Cae⁃norhabditis elegans(C.elegans)induced by high-salt and high-sugar diet.Methods C.elegans were cultured and ran⁃domly divided into the normal control group,high-salt group,high-sugar group and high-salt-high-sugar group.The nor⁃mal control group was added with nematode NGM medium.In the high-salt group,high-sugar group,and high-salt-highsugar group,we set up the model control group,10 mg/L berberine intervention group,and 20 mg/L berberine interven⁃tion group,which were first added with high-salt medium(nematode NGM medium containing 0.7%sodium chloride),high-sugar medium(nematode NGM medium containing 1%glucose),and high-salt-high-sugar medium(nematode NGM medium containing 0.7%sodium chloride and 1%glucose),respectively,and then the berberine intervention group was added with corresponding concentrations of berberine solution,and cultured for 6 days.The body length,body width of C.elegans were measured on the 1st,3rd and 6th days,the lifespan of C.elegans were measured and daf-2 and daf-16 mRNA levels were detected by RT-qPCR on the 6th day of culture.Results Compared with the normal control group,the high-salt,high-sugar and high-salt-high-sugar groups had significant increase in body width and significant reduction in lifespan(all P<0.05),while there was no significant change in body length(all P>0.05);the expression of daf-2 mRNA increased and daf-16 mRNA decreased(all P<0.05).Compared with the high-salt,high-sugar and high-salt-high-sugar groups,berberine supplementation significantly reduced body width and prolonged life span(all P<0.05),but did not af⁃fect body length(all P>0.05);the expression of daf-2 mRNA significantly decreased and daf-16 mRNA significantly increased(all P<0.05).Conclusion Berberine can reduce the morphological changes and lifespan shortening of C.ele⁃gans induced by high-salt and high-sugar diet probably by down-regulating the expression of daf-2 mRNA and up-regulating the expression level of daf-16 mRNA.
