不同厚度姜泥督灸对寒凝血瘀型痛经大鼠的治疗作用及机制研究 被引量:1

摘要 目的:探讨不同厚度姜泥督灸对寒凝血瘀型痛经大鼠的治疗作用及机制。方法:将36只健康雌性大鼠随机分为空白对照组、模型组及实验组,模型组及实验组构建寒凝血瘀型痛经模型,空白对照组仅腹腔注射等量生理盐水;造模成功后,将实验组分为2 mm组、3 mm组、4 mm组,各组给予相应厚度(2、3、4 mm)姜泥进行督灸,并腹腔注射盐酸替来他明盐酸唑拉西泮治疗,空白对照组、模型组仅给予腹腔注射等量盐酸替来他明盐酸唑拉西泮治疗。比较各组每分钟扭体评分、寒凝表征评分、全血还原黏度、丙二醛(MDA)及一氧化氮(NO)水平,观察实验组各亚组大鼠皮肤表现。结果:模型组、实验各组每分钟扭体评分、寒凝表征评分、全血还原黏度(中切、高切)、MDA水平高于空白对照组,NO水平低于空白对照组,差异有统计学意义(P<0.05);2 mm、3 mm、4 mm组每分钟扭体评分、寒凝表征评分低于模型组,3 mm、4 mm组全血还原黏度(中切、高切)低于模型组,3 mm、4 mm组NO水平高于模型组,3 mm组MDA水平低于模型组,差异有统计学意义(P<0.05)。2 mm组大鼠背部出现明显灸瘢,3 mm组大鼠灸后背部均出现短暂性红晕,部分出现轻微水疱,4 mm组大鼠灸后皮肤无明显异常。结论:督灸可改善寒凝血瘀型大鼠痛经症状,降低全血还原黏度、MDA水平,提高NO水平;选择姜泥厚度以皮肤出现红晕或轻微水疱疗效最佳。 Objective:To explore the treatment effect and mechanism of governor vessel moxibustion by different thicknesses ofginger paste in dysmenorrhea rats with cold coagulation and blood stasis.Methods:A total of 36 healthy female rats were randomlydivided into blank control group,model group and experimental group.In the model group and experimental group,rats were usedto establish the model of dysmenorrhea with cold coagulation and blood stasis.In the blank control group,rats were onlyintraperitoneally injected with equal amount of normal saline.After successful molding,the experimental group was divided into 2mm subgroup,3 mm subgroup and 4 mm subgroup,and each subgroup was given ginger paste at corresponding thickness(2 mm,3mm,4 mm) for moxibustion,and received intraperitoneal injection of Shutai.In the blank control group and model group,rats wereonly intraperitoneally injected with equal amount of Shutai.The score of writhing per minute,cold coagulation characterizationscore,whole blood reduced viscosity,malondialdehyde(MDA) and nitric oxide(NO) levels were compared between the groups.Theskin performance of rats in each subgroup of the experimental group was observed.Results:The score of writhing per minute,coldcoagulation characterization score,whole blood reduced viscosity(medium shear and high shear) and MDA level in the modelgroup and each experimental subgroup were higher than those in the blank control group,while NO level in the model group andeach experimental subgroup was lower than that in the blank control group,and the difference was statistically significant(P<0.05).The score of writhing per minute and cold coagulation characterization score in the 2 mm,3 mm and 4 mm subgroups werelower than those in the model group,the whole blood reduced viscosity(medium shear and high shear) in the 3 mm and 4 mmsubgroups was lower than that in the model group,the NO level in the 3 mm and 4 mm subgroups was higher than that in the modelgroup,and the MDA level in the 3 mm subgroup was lower than that in the model group,and the difference was statisticallysignificant(P<0.05).The rats in 2 mm subgroup had obvious moxibustion scar on the back,the rats in the 3 mm subgroup hadtransient flush and some slight blisters on the back after moxibustion,and there were no obvious abnormalities in the skin of the rats in the 4 mm subgroup.Conclusion:Governor vessel moxibustion can improve the symptoms of dysmenorrhea rats with coldcoagulation and blood stasis,reduce the whole blood reduced viscosity and MDA level,and increase the NO level.The thickness ofginger paste for the best results when the skin flushes or appear slight blisters.
