绿梅香橼双菌发酵滤液对皮肤屏障的影响及其机理研究

摘要 通过CCK8实验研究绿梅香橼双菌发酵滤液(Green plum/citron double-strain fermentation filtrate,DSF)对角质形成细胞(HaCaT)增殖率的影响,q PCR研究DSF对十二烷基硫酸钠(SLS)模型中HaCaT细胞丝聚合蛋白(FLG)、兜甲蛋白(LOR)、封闭蛋白1(CLD1)的mRNA转录水平的影响,Elisa实验研究DSF对模型中HaCaT细胞上清液中TNF-α含量的影响。结果表明,DSF对HaCaT细胞没有毒性,能提升模型中HaCaT细胞中FLG、LOR、CLD1的mRNA转录水平,能降低模型中HaCaT细胞上清液中TNF-α含量,DSF具有一定的强化皮肤屏障的护肤功效。 The effects of green plum/citron double-strain fermentation filtrate(DSF)on the proliferation rate of keratinocyte(HaCaT)were investigated by CCK8 assay.qPCR was used to study the effects of DSF on the mRNA transcription levels of HaCaT filaggrin(FLG),loricrin(LOR)and claudin1(CLD1)in sodium lauryl sulfate(SLS)model.The influence of DSF on the content of TNF-αin the supernatant of HaCaT cells was studied by ELISA.The results showed that DSF had no toxicity to HaCaT cells,and could increase the mRNA transcription levels of FLG,LOR and CLD1 in HaCaT cells and reduce the content of TNF-αin the supernatant of HaCaT cells in the model.Therefore,DSF has a certain skin-care efficacy of strengthening the skin barrier.
