长江刀鲚FABP1基因遗传多态性及其与生长性状的相关性分析

摘要 为选育出具有优良生长性状的长江刀鲚(Coilia ectenes)新品系,通过对长江刀鲚FABP1基因cDNA和DNA序列进行克隆、扩增和多态性SNP位点的筛选,初步分析这些位点与其生长性状的相关性。基于长江刀鲚的转录组数据,利用基因克隆技术获取长江刀鲚FABP1基因的cDNA和DNA序列,然后利用直接测序法筛选获得该基因上的SNP位点并进行基因分型,最后利用最小二乘法分析基因多态性及其与刀鲚生长性状的相关性。结果显示,该基因cDNA全长525 bp,开放式阅读框(ORF)由384个核苷酸组成,编码127个氨基酸;基因组DNA全长1 285 bp,包括4个外显子和3个内含子;共检测到14个SNP位点,分布于第1、2、3内含子和第3外显子上;14个位点的不同基因型与长江刀鲚的体长、体高和体质量性状没有显著的相关性(P>0.05);14个位点对长江刀鲚养殖群体遗传分析显示,刀鲚养殖群体的平均观测杂合度(Ho)、平均期望杂合度(He)和平均多态信息含量(PIC)分别为0.476、0.436和0.348。结果表明,长江刀鲚FABP1基因上的14个SNP位点虽然与其生长性状没有显著相关性,但是能够应用于长江刀鲚养殖群体的种质资源评估和分子标记辅助育种研究。 To study the correlation between genetic polymorphisms of CeFABP1 gene(FABP1 gene of Coilia ectenes)with the growth traits of C.ectenes,we cloned the cDNA and DNA sequence of CeFABP1 and screened the SNPs in this gene.Based on the transcriptome data of C.ectenes and gene cloning technology,we obtained the cDNA and DNA sequence of CeFABP1.The SNPs of CeFABP1 was screened and genotyped by DNA direct sequencing method,and the relationship between SNPs and growth traits of C.ectenes was analyzed by least squares method.The cDNA length of CeFABP1 was 525 bp,containing an open reading frame of 384 bp.The genomic DNA of CeFABP1 was 1285 bp in length containing three introns and four exons.Fourteen polymorphic sites were detected in intron 1(1),intron 2(11),intron 3(1)and exon 3(1),and were not significantly associated with all three growth traits of C.ectenes(P>0.05).The results of population genetic analysis of cultured population of C.ectenes based on the fourteen SNPs showed that the average values of observed heterozygosity(H_(o)),expected heterozygosity(He)and polymorphism information content(PIC)were 0.476,0.436 and 0.348,respectively.The data obtained herein provide useful information for further studies on the evaluation of germplasm resources and molecular marker assisted breeding in C.ectenes.
